P21-activated kinase in inflammatory and cardiovascular disease

Abstract: P-21 activated kinases, or PAKs, are serine–threonine kinases that serve a role in diverse biological functions and organ system diseases. Although PAK signaling has been the focus of many investigations, still our understanding of the role of PAK in inflammation is incomplete. This review consolidates what is known about PAK1 across several cell types, highlighting the role of PAK1 and PAK2 in inflammation in relation to NADPH oxidase activation. This review explores the physiological functions of PAK during … Show more

“…The downregulation of miR‐542‐5p has previously shown to be linked with advanced lymphatic metastasis, vascular invasion, TNM stage and EGFR, and survival analyses have widely highlighted that patients with lower miR‐542‐5p expression had poorer prognosis . P21‐activated kinases represent central regulators of the inflammatory response . PAK1 regulates the molecular mechanism of inflammation by the activation of its downstream target nicotinamide adenine dinucleotide phosphate oxidase in neutrophils .…”

“…Both Group I and II PAKs are important regulators of various biological processes including cell motility, survival, proliferation, and cytoskeletal organization. Owing to their involvement in the above physiological processes, deregulation of functional PAKs are implicated in multiple inflammatory disorders such as diabetes, cancer, mental retardation, allergy, inflammatory, and cardiovascular diseases ( 85 ).…”

“…Previous studies have reported that PAK1 activation was responsible for initiation of inflammation [22]. Khare et al found that overexpression of PAK1 promoted cell survival in inflammatory bowel diseases and Western blot in human OA chondrocytes transfected with siRNA-NC or siRNA-PAK1, followed by treating with 10 ng/ml IL-1β for 24 h; n = 3, **P < 0.01.…”
