Oxytocin Signaling Pathway: From Cell Biology to Clinical Implications

Abstract: Background: In addition to the well known role played on lactation and parturition, Oxytocin (OT) and OT receptor (OTR) are involved in many other aspects such as the control of maternal and social behavior, the regulation of the growth of the neocortex, the maintenance of blood supply to the cortex, the stimulation of limbic olfactory area to mother-infant recognition bond, and the modulation of the autonomic nervous system via vagal pathway. Moreover, OT and OTR show anti-inflammatory, anti-oxidant, anti-pa… Show more

“…In mammals, the two peptides have four cognate receptor subtypes encoded by four distinct genes: AVPR1A, AVPR1B, AVPR2 and OXTR. The actions of OT are mediated by the OT receptor (OTR) and include both behavioral and gonadal regulation and, in mammals, the powerful contractions of the uterus as well as stimulation of milk ejection (6). The responses to VT are triggered in mammals by three distinct receptor subtypes (7) and are more complex [see (8), for review].…”

“…Recent evidence indicates that perturbations of the thyroid hormone signaling pathway and antibodies are associated with GDM development and mal-outcome [ 49 , 51 ]. Emerging evidence suggests a role of OT in the pathogenesis of insulin resistance, obesity, and dyslipidemia [ 52 , 53 ], which are hallmarks or risk factors for GDM. A significant weight loss and improvement of insulin sensitivity, pancreatic β-cell responsivity, and lipid metabolism can be observed in rodents, nonhuman primates, and humans after chronic subcutaneous or intranasal OT treatment [ 54 , 55 ].…”

“…Collectively, Rh and AZ enriched 5 new pathways after their combined use. Among them, the oxytocin signaling pathway is related to the cardiovascular system (36,37). Oxytocin could reduce the force and rate of heart contraction and increase vasodilatation by mediating the atrial natriuretic peptide-Cyclic GMP (ANP-cGMP), and Nitric oxide-Cyclic GMP (NO-cGMP) pathways (38)(39)(40), which may reduce symptoms such as pulmonary hypertension or hypertension caused by AS (41).…”
