Cu-catalyzed 1,3-dipolar cycloaddition of ethyl 2-azidoacetate to iodobuta-1,3-diynes and subsequent Sonogashira cross-coupling were used to synthesize a large series of new triazole-based push–pull chromophores: 4,5-bis(arylethynyl)-1H-1,2,3-triazoles. The study of their optical properties revealed that all molecules have fluorescence properties, the Stokes shift values of which exceed 150 nm. The fluorescent properties of triazoles are easily adjustable depending on the nature of the substituents attached to aryl rings of the arylethynyl moieties at the C4 and C5 atoms of the triazole core. The possibility of 4,5-bis(arylethynyl)-1,2,3-triazoles’ application for labeling was demonstrated using proteins and the HEK293 cell line. The results of an MTT test on two distinct cell lines, HEK293 and HeLa, revealed the low cytotoxicity of 4,5-bis(arylethynyl)triazoles, which makes them promising fluorescent tags for labeling and tracking biomolecules.

The present work aims to evaluate Rosa x damascena Herrm. and Pelargonium graveolens L’Hér. essential oils, and the major constituent geraniol for their in vitro and in silico inhibitory activities against 5-lipoxygenase (5-LOX), cyclooxygenase (COX), acetyl cholinesterase (AChE), butyryl cholinesterase (BuChE), and angiotensin converting enzyme (ACE2) enzymes. Geraniol most potently inhibited the ACE2 relative to other enzymes. R. damascena essential oil moderately inhibited the cancer cell lines with no toxic effects on healthy HEK 293 cells. P. graveolens essential oil inhibited a number of cancer cell lines including A549, MCF7, PC3, and HEK 293 that are reported here for the first time. The molecular docking of geraniol with the target enzymes revealed that it binds to the active sites similar to that of known drugs. Geraniol carries the potential for further drug development due to its drug-like binding mode for the target enzymes. Our work confirms that these essential oils possess similar biological activities due to their similar phytochemistry in terms of the major constituents of the plants. The promising biological activities reported in this work further warrant the inclusion of in vivo studies to establish safe use of the target essential oils and their constituents.

Potential of Natural Antioxidants for Upregulating Apoptosis in Cervical Cancer Cells: A Cellular and Molecular Study

Natural antioxidants (NAs) are promising substances, which are established as potent therapeutic agents. However, the anticancer potential of many of them is not completely known. In this research, the anticancer effect of three natural antioxidants including geraniol (Ge), citronellol (Cit), and quercetin (Qu), and their combination by estimating their cytotoxicity on the Hela cells at doses from 15.625 to 1000 µg/mL was investigated. Moreover, cisplatin (Cis) was employed as a strong competitive comparison. After estimating the IC50 value of each treatment in the MTT assay, western blot was performed with the obtained doses for further evaluation. In this regard, the expressions of caspase3, Bax, Bcl2, and p53 proteins were examined. Our results showed that the combination of these natural substances had significant cytotoxic potential by upregulating all mentioned proteins, which demonstrated the apoptosis pathway. Our findings had also the same results in both Cis-receiving and non-Cis-receiving cells regarding some indices. Consequently, it was suggested to further investigate these natural antioxidants as both main or adjuvant therapies for their anticancer potential and side-effect reduction in vivo. Overall, it can be concluded that these natural antioxidants are effective against cancer cells by upregulating the apoptosis pathway.
