Oxidative stress and repetitive element methylation changes in artisanal gold miners occupationally exposed to mercury

Abstract: Mercury (Hg) exposure is a public health concern due to its persistence in the environment and its high toxicity. Such toxicity has been associated with the generation of oxidative stress in occupationally exposed subjects, such as artisanal gold miners. In this study, we characterize occupational exposure to Hg by measuring blood, urine and hair levels, and investigate oxidative stress and DNA methylation associated with gold mining. To do this, samples from 53 miners and 36 controls were assessed. We show hi… Show more

“…fact that "GSH replacing" antioxidants, such as of alpha-tocopherol acetate, N-acetyl cysteine, methionine, SAM, and folic acid methyl group donors also reversed the phenomenon of LINE-1 hypomethylation [49]. Since oxidative stress induces hypermethylation of the repetitive LINE-1 elements of SOD enzyme family and GSTM5 promoters [50,51] as well, and these positive feedback mechanisms have been disrupted by the substances studied in our model. DNA methylation and histone methylation are epigenetic factors that play a central role in tumor biology and are involved in the antitumor effects of polyphenols (e.g.…”

“…Additionally, LINE1 methylation was positively correlated with 8-hydroxy-2-deoxyguanosine(8-OHdG) and Hg levels. (24) These findings emphasize the negative effect of Hg on oxidative stress and DNA methylation in gold miners. Various studies (25, 87, 88) have reported alterations in membrane permeability as well as changes in macromolecular structure due to the high affinity of cellular mercury for sulfhydryl and thiol groups leading to DNA damage.…”

“…There is now some evidence that reactive oxygen species may induce changes in LINE-1 methylation. For example, in gold miners, oxidative stress associated with increased mercury levels, is associated with increased LINE-1 methylation (Narvaez et al, 2017). LINE-1 methylation is reduced in cells treated with peroxides (Kloypan, Srisa-art, Mutirangura, & Boonla, 2015), while antioxidants, including alpha-tocopherol, N-acetyl cysteine and NAC, Sadenosylmethionine and folic acid, may restore LINE-1 hypomethylation.…”
