ROS homeostasis is crucial to maintain radical levels in a dynamic equilibrium within physiological ranges. Therefore, ROS quantification in seeds with different germination performance may represent a useful tool to predict the efficiency of common methods to enhance seed vigor, such as priming treatments, which are still largely empirical. In the present study, ROS levels were investigated in an experimental system composed of hydroprimed and heat-shocked seeds, thus comparing materials with improved or damaged germination potential. A preliminary phenotypic analysis of germination parameters and seedling growth allowed the selection of the best-per-forming priming protocols for species like soybean, tomato, and wheat, having relevant agroeconomic value. ROS levels were quantified by using two noninvasive assays, namely dichloro-dihydro-fluorescein diacetate (DCFH-DA) and ferrous oxidation-xylenol orange (FOX-1). qRT-PCR was used to assess the expression of genes encoding enzymes involved in ROS production (respiratory burst oxidase homolog family, RBOH) and scavenging (catalase, superoxide dismutase, and peroxidases). The correlation analyses between ROS levels and gene expression data suggest a possible use of these indicators as noninvasive approaches to evaluate seed quality. These findings are relevant given the centrality of seed quality for crop production and the potential of seed priming in sustainable agricultural practices.

Para a obtenção de híbridos, um estudo inicial feito em linhagens de milho é primordial. Na seleção das linhagens, são observados diversos parâmetros para se chegar ao cruzamento e comercialização. Para aumentar a produtividade do milho, alguns autores afirmam ser necessária uma adubação nitrogenada adequada, sendo o N o macronutriente mais requerido pela cultura. Desempenha várias funções metabólicos nas sementes, contribuindo para sua formação e para sua reserva nutricional. Este trabalho teve como objetivo avaliar a produção e a qualidade de sementes de linhagem de milho irrigado em função de doses de nitrogênio e graus de umidade na colheita. O experimento foi conduzido na área experimental do IF Goiano - Campus Ceres e no LAS. Utilizou-se o delineamento experimental em blocos casualizados, sendo cinco doses de nitrogênio (50, 110, 170, 230 e 290 kg ha -1) e colheita com quatro graus de umidade (15, 25, 35 e 45%). Foi utilizada a linhagem de milho PGAG10051L. Realizaram-se testes de campo e laboratório. A linhagem estudada tolera baixa disponibilidade de nitrogênio, tendo alta eficiência no seu uso e responde positivamente ao aumento de nitrogênio quanto ao percentual de germinação, com melhores resultados acima de 170 kg N ha-1. A linhagem tem alta capacidade de acúmulo de biomassa nas cariopses mesmo com baixo teor de umidade. Sementes colhidas com 25% de umidade apresentaram resultados satisfatórios de germinação e vigor. Recomenda-se aplicar a dose 170 kg ha-1 de nitrogênio parcelada em quatro vezes para adubação em cobertura da linhagem de milho estudada.

Leaf stage as a defoliation criterion for the production of high-vigour annual ryegrass seeds

Previous research demonstrated that the interval between defoliation treatments based on the leaf stage is a promising tool for the management of annual ryegrass (Lolium multiflorum L.) pasture. The use of this tool, which is based on the physiological status of the plant, could also benefit the seed production. We assessed the effects of the time of defoliation based on leaf stage on seed yield and quality of annual ryegrass cut to three residual stubble heights (RSH) (50%, 25% and 0% of plant height pre-defoliation) and a second defoliation applied to certain treatments based on the leaf stage of the plant regrowth (two or three leaves per tiller). Higher defoliation intensities (lower RSH) increased the amount of harvested forage to 4.0 t DM ha–1. Moderate defoliation (50%) undertaken at the 3-leaf stage combined with intense defoliation (25% and 0%) at the 2-leaf stage resulted in the highest seed yields (1.8 t ha–1). However, lower RSH reduced mean seed weight and physiological seed quality, which was also associated with lower seed starch and protein content. Accelerated ageing (AA) tests were used to compare seed vigour between treatments. A second defoliation at the 3-leaf stage plus intense defoliation (RSH 0%) resulted in seed lots with a high rate of lipid peroxidation after the AA test, followed by lower catalase activity, germination, and seedling growth. The combination of a second defoliation at the 3-leaf stage and the retention of higher RSH (50%) promoted a greater proportion of leaf blades in the stubble, with a strong positive correlation with seed production and vigour. Antioxidant enzyme activity after the AA test is an important marker of annual ryegrass seed quality.
