Oxidative balance score and risk for incident prostate cancer in a prospective U.S. cohort study

Abstract: Purpose Oxidative stress is defined as an imbalance between pro-oxidants and antioxidants. Previous research found that a single comprehensive oxidative balance score (OBS) that includes individual pro-and anti-oxidant exposures may be associated with various conditions (including prostate cancer) in the absence of associations with the individual factors. We investigated an OBS-incident prostate cancer association among 43,325 men in the Cancer Prevention Study II Nutrition Cohort. Methods From 1999–2007, 3… Show more

“…Our findings regarding the overall OBS‐adenoma association are consistent with those reported in previous epidemiologic studies, which found inverse associations of oxidative balance scores (OBS) with colorectal adenoma, prostate cancer, and total cancer mortality . Of direct relevance to the present study, in an analysis of three pooled case‐control studies of incident sporadic colorectal adenoma ( n = 789 incident adenoma cases and 1,500 polyp‐free controls), the odds ratio comparing those in the highest relative to the lowest OBS tertile was 0.54 (95%CI 0.43‐0.69; p trend < 0.0001) .…”

“…48 OBS, measured by FFQs, has been used in a number of peer-reviewed publications. 16, 17, 19, 39 Similar associations were observed when OBS was measured by replacing questionnaire responses with biomarkers of pro- and antioxidant exposures. 37 …”

“…The purpose of OBS is to examine the balance of these modifiable factors; this approach has been used in a number of previous publications. 16, 17, 19, 39 In addition to extrinsic modifiable factors, oxidative stress is also influenced by intrinsic factors such as antioxidant enzymes and cellular energy balance; 49 These factors are not included in the OBS. Further, oxidative stress itself can be measured using a variety of biomarkers.…”

“…It is important to point out, however, that equal weighting approach is difficult to justify [20, 21]. Some previous studies assigned OBS weights based on the reported associations between individual OBS components and outcomes of interest such as colorectal tumors [22] and prostate cancer [23]; however no previous study used weighting based on biochemical measures of oxidative stress.…”
