Oxidative and Conjugative Metabolism of p–Nitroanisole and p–Nitrophenol in Isolated Rat Liver Cells

Abstract: Isolated rat liver cells were used in the study of p–nitroanisole and p–nitrophenol metabolism. p–Nitroanisole was o–dealkylated to form p–nitrophenol which was subsequently conjugated to form predominantly sulphate esters and β–glucuronides. 1. At low concentrations of p–nitrophenol, sulphate conjugation was predominant but with increasing substrate concentration the glucuronidation activity was increased. At 250 μM p–nitrophenol the major conjugate formed was glucuronide while sulphate conjugation was inhibi… Show more

“…74 Rotenone appeared to inhibit synthesis of glucuronidationreaction components in rat hepatocytes. 81 However, glucuronidase did not impact the polar metabolites that were observed in faeces from rotenone-treated rats. 67 Efforts are beginning to be made to understand the contributions of gut microbiota to drug metabolism.…”

“…In vertebrates, an early paper suggested an additional, possible, glucuronidation step that yielded water-soluble metabolites . Rotenone appeared to inhibit synthesis of glucuronidation-reaction components in rat hepatocytes . “…Conjugation of 4-nitrophenol also occurred in cultured skin epithelial cells from humans (Rugstad and Dybing 1975), in isolated rat hepatocytes (Araya et al 1986;Moldeus et al 1976;Tonda and Hirata 1983), and in microsomes isolated from dog livers (Nakano et al 1986).…”

“…Cytosolic SULT1A1 activity was determined by measuring the sulfoconjugation of p-nitrophenol (Campbell et al, 1987;Moldeus et al, 1976). Incubations were conducted using 10 mg/mL cytosolic protein, 1 mM 3′-phosphoadenosine-5′-phosphosulfate (PAPS) as a sulfate donor, 5 mM p-nitrophenol, in 50 mM Tris buffer pH 7.4, containing 10 mM magnesium chloride, 0.1 mM EDTA in a total volume of 100 μL.…”
