Overexpression of OgPAE1 from wild rice confers fungal resistance against Botrytis cinerea

Abstract: A full-length cDNA of the OgPAE1 gene encoding the alpha5 subunit of the 20S proteasome was isolated from wild rice (Oryza grandiglumis) treated by wounding or with a fungal elicitor. The deduced amino acid sequence of OgPAE1 comprises 237 amino acids (25.99 kDa), and shows 94.5% homology with Arabidopsis thaliana AtPAE1. Expression of OgPAE1 is regulated by defense-related signaling chemicals such as cantharidin, endothall and jasmonic acid. Overexpression of OgPAE1 in A. thaliana leads to resistance to the f… Show more

“…The expression of the α 5 subunit from Oryza grandiglumis (OgPAE1) is induced during defence responses, and the overexpression of OgPAE1 in A. thaliana leads to resistance against Bo. cinerea , but the underlying mechanism(s) remains unknown (Jeon et al. , 2008).…”

“…The expression of the a5 subunit from Oryza grandiglumis (OgPAE1) is induced during defence responses, and the overexpression of OgPAE1 in A. thaliana leads to resistance against Bo. cinerea, but the underlying mechanism(s) remains unknown (Jeon et al, 2008). The implication of the proteasome a5 subunit in plant defence might be more important in resistance to viruses, as it has been shown to specifically cleave in vitro TMV RNA Petit et al, 1997).…”

“…Fully expanded wild rice leaves were cut in approximately 2-cm-long segments, and floated on solution in sterile petri dishes. The tissues were treated with either distilled water for the control or 20% yeast extracts, wounding or various chemicals; 100 lM CN (Cantharidin), 100 lM EN (endothall), 1 mM ET (ethephon), 100 lM JA (jasmonic acid) or 100 lM SA (salicylic acid) (Jeon et al 2008;Rakwal et al 2001). Treated samples were harvested at the indicated times, and used immediately for RNA isolation or stored at -80°C.…”

“…This species has large panicles and has potentially useful genes for many agronomic traits such as higher biomass and resistance to diseases and insect (Heinrichs et al 1985;Yu et al 1997;Vaughan et al 2003). A few studies have been conducted to detect genetic variability of O. grandiglumis (Shin et al 2004;Kim et al 2005;Jeon et al 2008Jeon et al , 2009Pak et al 2009;Jeon et al 2012;Shin et al 2012). However, attempts to transfer genes from O. grandiglumis to cultivars have been limited mainly due to incompatibility between unrelated genomes (Yu et al 1997).…”
