Overexpression of homogentisate phytyltransferase (HPT) and tocopherol cyclase (TC) enhances α-tocopherol content in transgenic tobacco

Abstract: Photosynthetic organisms synthesize the amphipathic antioxidants called tocopherols which are essential components of the human diet. To increase the α-tocopherol (vitamin E) content, Arabidopsis genes encoding homogentisate phytyltransferase (HPT) and tocopherol cyclase (TC) were constitutively expressed individually and in combination (HPT:TC) in tobacco plant by Agrobacterium mediated transformation. The transgene was confirmed by polymerase chain reaction (PCR), transgene expression was studied by reverse … Show more

“…The overexpression of HPT and TC may cause tocopherol accumulation during plant growth. Harish and others found that the α-tocopherol content in transgenic tobacco plants was increased by 5.4-, 4.0-, and 7.1 folds with expressions of HPT , TC , and HPT:TC , respectively, when compared to the wild type [ 24 ]. Our results showed an agreement with this rule.…”

“…Leucoanthocyanidin dioxygenase was involved in biosynthesis of proanthocyanidin and vacuole development [ 54 ]. The overexpression of homogentisate phytyltransferase, considered as a key enzyme limiting tocopherol biosynthesis in lettuce, improved the content of tocopherol, a lipid soluble compound functioning as antioxidants and involved in signal transduction and transcription regulation [ 55 ]. Cytochrome P450 (CYP450) characterized as heme-thiolate enzymes was necessary for carbon assimilation, synthesis of hormones and as a structural components of cells [ 56 ].…”

“…Previous studies reported that overexpression of the Arabidopsis VTE1 gene in seeds of transgenic rapeseed plants, and in leaves of Arabidopsis , transgenic tobacco and transgenic lettuce, all resulted in an increase in total tocopherol, due mainly to an increase in γ-tocopherol (Kumar et al 2005 ; Kanwischer et al 2005 ; Lee et al 2007 ; Yabuta et al 2013 ; Harish et al 2013 ). In addition, overexpressing VTE1 can enhance tolerance to the environmental stresses.…”
