Overexpression of C/EBPβ Affects The Cell Cycle Regulators and Spermatogenesis Related Genes Expression And Function of Bovine Sertoli Cells

Abstract: CCAAT/enhancer-binding protein beta (C/EBPβ), an important transcriptional factor, plays a pivotal role in the regulation of female germ cell development. However, the role of C/EBPβ on the development of male germ cells has not been reported. In this study, we constructed the recombinant adenovirus plasmids of bovine C/EBPβ gene and harvested the subsequent adenoviruses, and then assessed the mRNA levels of spermatogenesis-related genes (real-time PCR) and secretion of inhibin B after 48 h of Ad-C/EBPβ recomb… Show more

“…Sertoli cell proliferation under diverse stimuli has been correlated with the levels of cyclins D1, D2, and E1 in different experimental models. [31][32][33][34][35][36] Moreover, p21 Cip and p27 Kip expressions have been associated with the detention of proliferation and with the increase in the levels of differentiation markers of Sertoli cells. 18,30,37 Results obtained herein show that the decreased proliferation caused by RSV treatment in 8-day-old Sertoli cells is accompanied by decreased expression of cyclins D1, D2, E1, and E2 and increased expression of p21 Cip and p27 Kip , suggesting that the observed effect of RSV on proliferation under basal conditions may be explained, at least in part, by the variation of these cell cycle regulators.…”

“…The differential adherent selection method was used to isolate immature bovine SCs from 6-week-old Holstein calves as described by Yang et al (2014) and Tang, Jin, Chen, and Wang (2016). The purity of a primary SCs culture was identified using the Leydig cells marker 3β-HSD and the SCs marker androgen binding protein (ABP) (Harikae, Tsunekawa, Hiramatsu, Toda, & Kurohmaru, 2012).…”

“…Primary Sertoli cells (PSC) were isolated from the testes of neonatal Qinchuan bulls (n = 3) and cultured as previously described (Yang et al, 2014;Tang et al, 2016). Briefly, the testes were decapsulated and digested in 1.0 mg/mL collagenase IV/DNase solution (Sigma, USA), and incubated at 37 °C in a humid environment containing 5% CO 2 until the tubules were separated (about 45 min).…”
