Osteoprotegerin Inhibits Vascular Calcification Without Affecting Atherosclerosis in ldlr (−/−) Mice

Abstract: Background-The role of osteoprotegerin in vascular disease is unclear. Recent observational studies show that serum osteoprotegerin levels are associated with the severity and progression of coronary artery disease, atherosclerosis, and vascular calcification in patients. However, genetic and treatment studies in mice suggest that osteoprotegerin may protect against vascular calcification. Methods and Results-To test whether osteoprotegerin induces or prevents vascular disease, we treated atherogenic diet-fed … Show more

“…Calcification commonly complicates longstanding atherosclerosis, and the absence of OPG in OPG/apoE double knockout mice accelerates calcific atherosclerosis that develops in apoE−/− mice (75). These findings suggest that OPG could also limit calcification of atherosclerotic plaques (76). However, whether OPG or RANKL play important roles in cardiovascular disease remains to be determined and is controversial (77).…”

“…In this study, CKD together with high-fat diet also increased both intimal and medial calcification. In another study of LDR Ϫ/Ϫ mice that were fed an atherogenic diet, treatment with recombinant osteoprotegerin reduced the calcification area and the expression of osteocalcin, a marker of mineralization, without affecting size or number of aortic atherosclerotic lesions (54). In an animal model of type 2 diabetes, TNF-␣ was shown to induce the MSX-Wnt osteogenic program that regulates arterial calcification (32).…”

“…Real-time reverse-transcriptase polymerase chain reaction (RT-qPCR) was performed using the One-Step qRT-PCR SuperMix Kit (BioChain Institute, Inc., Hayward, CA, USA) and Mx3005P Real-Time PCR System (Agilent Technologies, Inc., Wilmington, DE, USA), as described previously. (27)(28)(29) Statistical analysis HFD effects on gene expression and bone parameters were compared in chow versus HFD groups by Student's t test. Comparisons across more than two groups were performed with two-way ANOVA, followed by the Fisher's PLSD test using…”
