Osmolality and composition of the extender during the cold storage of Prochilodus lineatus (Characiformes: Prochilodontidae) sperm

Abstract: This study evaluates the effects of extender osmolality and composition on the cooling of Prochilodus lineatus sperm. Sperm was diluted in six extenders: two compositions (powdered coconut water™ = ACP; Beltsville Thawing Solution™ = BTS) x three osmolalities (285, 325, and 365 mOsm/kg) plus an undiluted control, and stored at 6-8°C. Motility rate and velocities (curvilinear, straight line, and average path) were determined every other day. Osmolality did not affect the quality of cooled sperm, thus data were … Show more

“…Semen analysis using CASA has been reported in other species of fish and has been shown to be a good tool to help determine sperm quality; it provides more information than subjective analysis and more accurate results (NYNCA et al, 2014;VIVEIROS et al, 2014;JUDYCKA et al, 2015;MARIA et al, 2015). This is one of the first studies using CASA in the analysis of fresh and cryopreserved sperm for P. brevis.…”

“…The extender solution powdered coconut water ACP (ACP Biotecnologia, Fortaleza, CE, Brazil), was developed for several freshwater species; this solution has recently been adapted for semen fish, received the name ACP-104 (Viveiros et al, 2010), and was tested in Prochilodus lineatus (Viveiros et al, 2014b).…”

“…Fish semen cryopreservation technology has expanded considerably worldwide with the use of portable nitrogen vapour cylinders, known as “dry‐shippers.” Semen is packaged in straws, added to the cylinder and subjected to uncontrolled speeds between 30 and 40°C/min. Fish semen from neotropical Characiformes and Siluriformes orders has been successfully frozen using this technique (Viveiros, Taffarel, & Leal, ).…”
