Ortho-phthalic acid esters in lipophilic extract from the cell culture of Aconitum baicalense Turcz ex Rapaics 1907

Abstract: Optically active bis-2R(-)ethylhexyl o-phthalate was obtained with 0.18% yield from dry cultured cells of Aconitum baicalense Turcz ex Rapaics 1907 by extraction with petroleum ether followed by silica gel column chromatography. Its structure was confirmed by the analysis of C andH NMR spectra. Seasonal fluctuations of quantitative phthalate content in A. baicalense cells were identified. The tests were performed under conditions excluding the presence of phthalates in reagents, materials, and laboratory dishe… Show more

“…Although some Aconitum species are highly toxic because of aconite alkaloid, many species are essential in the formulation of traditional herbal medicine in Asia ( Zhao et al, 2010 ; Semenov et al, 2016 ; Wada et al, 2016 ; Liang et al, 2017 ). The current state of Aconitum phylogenetics lacks molecular information of some species in Eastern Asia, and thus inhibits identification and germplasm utilization of this genus.…”

“…At the same time, the measured stable carbon isotopes 13 C/ 12 C ratio in DEHP explains the phenomenon of its high concentration originated from a potential biotic source. It should be noted that (i) DEHP congener is a potential carcinogenic compound [39] and its MPC is high, equal to 8.0 µg/L; (ii) structurally, DEHP is presented by two stereoisomers, the only one of which, namely bis-2R(-)ethylhexyl phthalate, has been isolated from environmental objects so far; for example, the one isolated from the cells of the Aconitum baicalense (Turcz ex Rapaics 1907) culture and from brown sugar [40,41]; (iii) DEHP toxicological studies were conducted using a synthetic racemic product, while the natural DEHP stereoisomer does not have to obligatory possess the same physiological properties [42]; (iv) while obtaining extreme DEHP concentrations, it is necessary to identify the source which could be biogenic or anthropogenic, one or both, before taking any environment protections.…”

“…Measurement of carbon stable isotopes 13 С/ 12 С in DEHP explains a phenomenon of high concentrations of this congener by its income from biotic sources [23]. Due to this fact, we have to notice the following: i) congener DEHP is related to potential carcinogens [37] and MAC for it is "the severest"; ii) DEHP structure is presented by two stereoisomers but only one optically active bis-2R(-)ethylhexyl phthalate is extracted from environmental objects, for example, from a biomass of culturable cells of Aconitum baicalense (Turcz ex Rapaics 1907) and from brown sugar [38,39]; iii) toxicological studies of DEHP were done at a synthetic racemic product, and a natural stereoisomer of DEHP has not to possess mandatory the same physiological properties [40]; iv) while revealing extreme concentrations of congener DEHP, before taking due environment protection measures, it is necessary to identify a DEHP source -biogenic or anthropogenic one.…”
