Organogenesis and somatic embryogenesis in callus cultures ofRosa hybrida andRosa chinensis minima

Abstract: Pre-incubation of somatic tissues of the cut rose 'Carefree Beauty' and miniature roses 'Red Sunblaze' and 'Baby Katie' in 10, 100, or 200 #M 2,4-D induced the development of highly rhizogenic callus. Upon transfer ofrhizogenic callus to a regeneration medium containing 23 #M TDZ and 3 #M GA3, a low frequency of shoot organogenesis (3.3%) and somatic embryogenesis (6.6%) was observed. Incubation of leaf and stem internodes in 11, 27, 54, 81, or 108/zM NAA followed by transfer of explants to the regeneration me… Show more

“…Similar results were found on media containing sucrose and fructose. The superiority of glucose in promoting shoot regeneration from stem explants of P. amurense is consistent with the results obtained with Beech, Rosa hybrid and chinensis minima (Cuenca and Vieitez, 2000;Hsia and Korban, 1996;Lemos and Baker, 1998). This may be because that glucose is more efficiently uptake and transported in tissues or more immediately used to provide energy for shoot organogenesis.…”

“…In general, sucrose is commonly used in tissue culture media to induce adventitious shoots or buds. However, there are few reports whereby glucose has been found to be better source of carbon than sucrose (Hsia and Korban 1996, Sul and Korban 1998, Cuenca and Vietiez 2000. Among carbon sources tested, sucrose was found best for multiple shoot proliferation of banana.…”

“…These researchers observed also that the tissue producing roots diff erentiated shoots very rarely. On the other hand, in some plant species shoots regenerated only from callus with the high level of rhizogenesis (Hsia and Korban, 1996). Because auxin is gradually degraded on the light (Stasinopoulos and Hangarter, 1990), hence may arise a much higher degree of the rhizogenesis noted for the explants cultivated in the dark.…”
