Organization of the Haemophilus ducreyi 35000 chromosome

Abstract: A physical and rudimentary genetic map of the Haernophilus ducreyr' strain 35000 genome was constructed. Pulsed-f ield gel electrophoresis was used to separate restriction fragments of H. ducreyi DNA digested with Sfil, I-Ceul, or Sfil plus I-Ceul. The sizes of the fragments were determined, and the circular chromosome was estimated t o be 1757 kbp. The six I-Ceul fragments and four Sfil fragments were ordered into macrorestriction maps using Southern blot hybridization with random H. ducreyi clones as probes.… Show more

“…The sizes of the chromosomes of the A. pleuropneumoniae type and serotype reference strains were determined to range between 2.3 and 2.4 Mb using PFGE analysis of AscI and ApaI digested chromosomal DNA. The size of the A. pleuropneumoniae chromosome is comparable to that of other species of the family Pasteurellaceae which includes Actinobacillus actinomycetemcomitans (2.3 Mb) [21], Haemophilus parain£uenzae (2.34 Mb), Haemophilus in£uenzae (1.83 Mb) [22] and Haemophilus ducreyi (1.76 Mb) [23]. The sizes of the chromosomes of the di¡erent A. pleuropneumoniae strains analyzed vary relatively little compared to other bacterial species.…”

The sizes of the chromosomes of the different A. pleuropneumoniae strains analyzed vary relatively little compared to other bacterial species.…”

“…The rRNA genes and groEL gene exist in multiple copies in most eubacteria, including H. ducreyi 19,33 . Species-speci® c regions of these evolutionarily conserved genes have been successfully used as targets for PCR assays for the detection and differentiation of pathogenic microorganisms 31 .…”
