Chromosome sizes and phylogenetic relationships between serotypes of Actinobacillus pleuropneumoniae

Abstract: The genome size of Actinobacillus pleuropneumoniae was determined by pulsed field gel electrophoresis of AscI and ApaI digested chromosomal DNA. The genome size of the type strain 4074 (serotype 1) was determined to be 2404 þ 40 kb. The chromosome sizes for the reference strains of the other serotypes range between 2.3 and 2.4 Mb. “…After being washed with wash buffer [20 mM tris-HCl (pH 8), 50 mM EDTA and distilled water] and then digested with proteinase K solution overnight at 55°C, each genomic DNA was digested with AscI (New England Bio Labs Inc., Beverly, MA, USA) and ApaI (Takara Bio Inc., Otsu, Japan) for 18 hours.
