Oral myeloid cells uptake allergoids coupled to mannan driving Th1/Treg responses upon sublingual delivery in mice

Abstract: BackgroundPolymerized allergoids coupled to nonoxidized mannan (PM‐allergoids) may represent novel vaccines targeting dendritic cells (DCs). PM‐allergoids are better captured by DCs than native allergens and favor Th1/Treg cell responses upon subcutaneous injection. Herein we have studied in mice the in vivo immunogenicity of PM‐allergoids administered sublingually in comparison with native allergens.MethodsThree immunization protocols (4‐8 weeks long) were used in Balb/c mice. Serum antibody levels were teste… Show more

“…In the present study, we demonstrated that AIT differentially modifies the activation and function of several Treg subsets. AIT in vivo increased subsets of allergen‐specific Treg cells with distinct phenotypes, and the mechanisms of action include the CD4 + CD25 + CD127 − FOXP3 + nTreg and the CD4 + IL‐10‐secreting Tr1 cells . In addition to changes in allergen‐specific Treg cells, we observed that in 9 out of 12 patients, overall IL‐10 + Tr1 cells also increased during AIT, which is concordant with the previous data reported by others …”

“…Recent findings have shown that allergoids conjugated to nonoxidized mannan from Saccharomyces cerevisiae are next‐generation vaccines targeting DCs through CLRs. These vaccines are candidates for AIT of allergic diseases as they promote the generation of Treg cells by mechanisms partially depending on programmed death‐ligand 1 (PD‐1) and IL‐10 in both humans and mice . Phase II clinical trials for grass‐pollen and house dust mite allergens are currently ongoing …”

“…9,10 In mice, subcutaneous or sublingual immunization with PM increases the frequency of splenic FOXP3 + Treg cells and induces healthy immune responses to allergens. 9,11,13 All these beneficial effects depend on the structural integrity of the conjugated mannan. 9,[12][13][14] Aluminium hydroxide (alum) remains the most widely used adjuvant in AIT.…”
