Oral Absorption of Flucloxacillin in Infants and Young Children

Abstract: The bioavailability of flucloxacillin (Heracillin@) following oral administration was determined in infants and children. After a dose of 12.5 mg/kg, peak concentrations were achieved at 60 min. which declined rapidly and were low in older children only 4 hours afterwards. Infants below 6 months of age showed a better absorption when given the mixture than older children. Older children achieved higher plasma concentrations when given tablets than when given equal doses of the mixture. There was no difference … Show more

“…The administration of flucloxacillin oral liquid to children can be a challenge and parent/carers should be supported with this task. Previous knowledge of the impact of food on the bioavailability of flucloxacillin in children is not conclusive . Regular reports of mixing flucloxacillin with food highlights the need for additional research to understand the impact on pharmacokinetics of co‐administration with food to improve palatability rather than dosing with a full meal.…”

[18] Regular reports of mixing flucloxacillin with food highlights the need for additional research to understand the impact on pharmacokinetics of co-administration with food to improve palatability rather than dosing with a full meal. These data demonstrate the need for production of more palatable paediatric formulations of flucloxacillin.…”

“…Oral penicillin was characterized by a lower C max value compared with penicillin administered intramuscularly [ 68 ]. Studies revealed a better absorption of oral flucloxacillin in infants 0–1 month old compared with older children [ 69 ], especially for such formulations as mixtures [ 70 ], plasma concentrations after both intravenous (i.v.) and oral administration were well above the MIC -values generally reported for Staphylococcus aureus [ 71 ].…”

“…However, neonates had higher absorption than older children, 25 and infants aged under six months had better absorption than older children with liquid formulations. 26 Higher doses and weight-based doses that are recommended in many guidelines for bone infections in children are not included in the product information.…”
