Optimization of the Production of Lovastatin from Aspergillus sclerotiorum PSU- RSPG178 under Static Liquid Culture using Response Surface Methodology

Abstract: The effect of lovastatin on cholesterol synthesis is well known. Although lovastatin has been produced from several Aspergillus species and cultivation systems, the yield of lovastatin from Aspergillus sclerotiorum PSU-RSPG178 in a static liquid system has not yet been determined. Therefore, the objective of this study was to optimize lovastatin production from this strain using a central composite design developed through response surface methodology (RSM). The inoculum was prepared from mycelium or spore cul… Show more

“…Mycelial plugs (0.5 cm×0.5 cm) cut from the periphery of an actively growing colony were used as the inoculum according to the method of Daengrot et al (15). The mycelial plugs were used since a spore inoculum gave a low yield of lovastatin in a previous study (14).…”

“…In this context, the current world trend is the search for a bioprocess efficient and sustainable for the production of biomolecules with high added value, however, a major challenge is to find microorganisms with high enzymatic potential that allows the production of these biomolecules. The biotechnological production of the HMG-CoA reductase inhibitor gains prominence due to the possibilities of using renewable and low-cost sources for their achievement, contributing to the essential requirements of sustainability (Morofuji et al, 2022;Mahmoud;Abdel_Hadi, 2022;Kashour et al, 2021;Suwannarat et al, 2019;Mouafi;Ibrahim;Elsoud, 2016;Jaivel;Marimuthu, 2010).…”

“…A statistical experimental design grants an ordered and pro cient design to accomplish the targets. At present, Plackett-Burman design (PBD) and Response Surface Methodology (RSM) which is a central composite method (Suwannarat et al 2019) widely employed to determine the critical factors quickly from multi-variables (Bae and Shoda, 2005;Usha et al 2011;Vohra and Satyanarayana, 2002;Baskar and Renganathan, 2009;Vuddaraju et al 2010;Ramon-Portugal et al 1997). RSM is a compilation of statistical techniques applied for experimental designs, generating models, determining the in uence of factors and detecting the factors of optimum conditions for advantageous responses (Vohra and Satyanarayana, 2002).…”
