Optimization of tetrazolium tests to assess the quality of Platymiscium floribundum , Lonchocarpus muehlbergianus and Acacia polyphylla DC. Seeds

Abstract: -The tetrazolium (TZ) test is an alternative to conventional germination tests, which usually require longer periods for seed quality evaluation. However, it is necessary to develop specific methodologies for each species, determining the concentration and time of exposure to salt. Thus, the objectives of this study were (i) to optimize the application of tetrazolium in forest seeds of Platymiscium floribundum Vog., Lonchocarpus muehlbergianus Hassl. and Acacia polyphylla DC., with the proposal of an evaluatio… Show more

“…The methodological adjustment of the tetrazolium test for each species is necessary since seeds differ regarding their morphological characteristics and chemical compositions (Abbade & Takaki, 2014;Silva et al, 2016;Masullo et al, 2017). It is important to adapt a methodology to P. taeda, aiming to facilitate the exposure of tissues prior to staining and to define an appropriate period of staining, considering the different procedures recommended by Brasil (2009) and Baalbaki et al (2009).…”

“…This is also corroborated with the ISTA rules of tetrazolium test [13]. Masullo et al (2017) optimized the tetrazolium tests to assess the viability and quality of Platymiscium floribundum, Lonchocarpus muehlbergianus and Acacia polyphylla seeds for different concentration of tetrazolium solution, incubation time and temperature [32]. Thus, TZ solution of 0.5%, incubation at 25°C temperature for 24 hours standardized as an optimum condition for TZ staining in Pinus roxburghii seeds for the seed viability and quality testing.…”

“…Além da variável de Tzv, foram calculadas a porcentagem de sementes vigorosas (Tzvg), e o produto da viabilidade e vigor (Tzvvg) em nível de repetições obtido pela expressão Tzvvg = [(Tzv)(Tzvg)]/100 (GARCIA et al, 2020;MASULLO et al, 2017;BRASIL, 2009).…”
