Obesity and its associated diseases such as diabetes mellitus and coronary heart diseases are a major challenge for our society. An important target for the treatment of obesity includes the development of inhibitors of nutrient digestion and absorption. Inhibition of pancreatic lipase and the associated reduction of lipid absorption is an attractive approach for the discovery of potent agents. Currently, the only clinically approved pharmacologic agent as pancreatic lipase inhibitor is Orlistat. However, its usage is compromised by unpleasant gastrointestinal adverse reactions (oily stools, oily spotting, flatulence). The use of botanical materials as a potential source of new drugs is of increasing importance and application. Natural products that are interesting for obesity treatment are generally considered to have less toxic and side effects than totally synthetic drugs. One of the most important sources of potential pancreatic lipase inhibitors represents the class of polyphenols. This article summarizes most studied subclasses of polyphenols including flavonoids, hydroxycinnamic acids, hydroxybenzoic acids and lignans with pancreatic lipase inhibitory effects. A structural comparison of potent inhibitors shows an increased inhibitory effect depending on number and position of phenolic hydroxyl groups, degree of polymerization and elimination of glycosylation during digestion.

Efficient preparation of loganin from Cornus officinalis fruits was investigated. First, effect of extraction conditions on loganin yield was measured. The loganin content in C. officinalis extract was greatly affected by ethanol concentration and extraction time whereas extraction temperature exerted relatively little effect. Response surface methodology with Box-Behnken design suggested optimized extraction condition for maximum loganin yield as ethanol concentration, 32.0%; temperature 46.2 °C and extraction time, 46.7 min, which yielded 10.4 μg loganin/mg dried fruit. Next, the effect of maturation stage of C. officinalis fruits on loganin content was investigated. The loganin content in the extract of C. officinalis fruits was decreased as the maturation process. The loganin content in the unripe fruits was 18.0 μg/mg extract whereas reduced to 13.3 μg/mg extract for ripe fruits. Taken together, our present study suggested the importance of extraction condition and maturation stages for efficient preparation of loganin from C. officinalis fruits.

In this article, the effect of the solvents (water, methanol, ethanol, chloroform, N-butanol and ethyl acetate) on the extraction yields (EY), total flavonoids content (TFC), total phenols content (TPC) form walnut shell and the inhibitory effect of extracts on pancreatic lipase were analysed. The kinetics of enzyme inhibition and was investigated. The inhibitory effect of extracts on adipogenesis of 3T3-L1 preadipocytes was also investigated. The highest EY, TFC and TPC were achieved with N-butanol (4.54%), Ethyl acetate (80.40 mg QEs/g extract) and ethyl acetate (200.40 mg GAE/g extract). The order of inhibitory activity was methanol extract (ME) > ethyl acetate extract (EAE) > ethanol extract (EE) > water extract (WE) > N-butanol extract (NBE) > chloroform extract (CE), the inhibition ratios were 82.51%, 73.13%, 70.49%, 69.42%, 65.57% and 60.66%, respectively under the concentration of 500 μg/mL. The IC 50 values of WE, ME, EE, CE, NBE and EAE were 371.35, 308.91, 407.57, 196.50, 247.79 and 254.27 μg/mL respectively. The inhibitory types of walnut shell extracts were noncompetitive. All the extracts were non-toxic to 3T3-L1 preadipocytes. The ME, EE, CE, NBE and EAE have inhibitory effect on triglyceride accumulation of 3T3-L1 cells.
