Optimization of nanofibrous silk fibroin scaffold as a delivery system for bone marrow adherent cells: in vitro and in vivo studies

Abstract: Electrospun silk fibroin nanofibrous scaffolds (ESFNSs) were successfully prepared by electrospinning of various Bombyx mori silk fibroin concentrations (10, 12, and 14% in formic acid). After characterizing the purified silk fibroin, the morphology, porosity, fibers' diameter, and uniformity of the prepared scaffolds were examined in detail. In addition, biological responses such as effects on bone marrow cell viability, cytotoxicity, and cell adhesion were evaluated in vitro. Biocompatibility and bioactivity… Show more

“…Short‐term cell viability and cytotoxicity were measured by MTT (Gholipourmalekabadi et al, ) and LDH‐specific activity assay (Rostami et al, ), respectively. For this purpose, the cells were cultured on the scaffold for 24, 48, and 72 h as described in our previous studies (Johari et al, ; Johari et al, ; Kazemi et al, ) and their proliferation rate was compared with those of the cells cultured in a culture plate (as a positive control).…”

“…The MSCs were seeded on both HA/GEL and HA/GEL/OC nanocomposites and incubated for 3 days, as described in our last published work . Thereafter, the samples were rinsed with distilled water, fixed with gluteraldehyde 2.5% solution and dehydrated in a graded concentration (30, 50, 70, and 100%) of acetone (Merck, Germany).…”

“…The sections were stained with Harris Hematoxylin (EM Science) and eosin Y (Sigma, USA), and Malory's trichrome for histological analysis. 24,25 The rate of collagen synthesis, the presence of inflammatory cells and number of fibroblast in the defected area were the major criteria evaluated in this study.…”
