Optimization of genome shuffling for high-yield production of the antitumor deacetylmycoepoxydiene in an endophytic fungus of mangrove plants

Abstract: As an accelerated evolutionary tool, genome shuffling is largely dependent on the high fusion frequency of different parental protoplasts. However, it was unclear how many types of parental protoplasts would afford the highest fusion frequency. Here, we applied the Monte Carlo method to simulate the simplified processes of protoplast fusion, to achieve maximal useful fusions in genome shuffling. The basic principle of this simulation is that valid fusions would take place when the minimum distance between two … Show more

“…Despite the attractive features in structure and activity, MED research suffered from the low yield in strain A123. Subsequently, we used the genome shuffling approach to generate high MED-producing strains, which were screened by the high-throughput screening method, ‘Antimicrobial-TLC-HPLC’ (ATH) [ 16 , 17 ]. These efforts led to several high yield strains for MED analogs, including strain A818, in which the yield increased over 200-fold.…”

“…Deacetyl-mycoepoxydiene (DA-MED) is a 248 molecular weight compound that has been isolated from the endophytic fungus, Phomopsis sp., of costal mangrove plants and has been shown to be a secondary metabolite with a rare oxygen-bridged cyclopentadiene skeleton [51]. This compound has cytotoxic activities toward various cell lines, including A549, HCC-S102 and HepG2 cells with IC 50 values ranging from 1.05 to 1.95 mg/ml.…”

“…26 The first step in the construction of new hybrids is to create a parental library, using random mutagenesis and rapid screening. 27 This way, a parental library with strong resistance properties is created. These strains are then subjected to the protoplastization and further combined in the protoplast fusion.…”
