Optimization of extraction condition for major iridoid components in fruit of corni (Cornus officinalis) by UPLC-PDA using response surface methodology

Abstract: The fruit of corni (Cornus officinalis Sieb.) was analyzed by ultra-performance liquid chromatography with a photodiode array detector (UPLC-PDA) and method validation for 3 major components which are morroniside, loganin, and cornuside. The optimal extraction condition for sum of 3 components was carried out response surface methodology (RSM). Experimental factors were designated by RSM and a central composite design (CCD). The suitability of the experimental model was verified by analysis of variance (ANOVA)… Show more

“…In order to estimate the optimization of the UAE process of Sideritis raeseri , we use the RSM as a compilation of mathematical and statistical analyses established on the fit of polynomial equations to the experimental data [ 14 ]; this describes well the behaviour of the dataset, aiming to make statistical previsions; since it reduces time, space, and raw material usage, it is more favourable than the traditionally used single parameter optimization [ 15 ]. Such a model of the optimization of the UAE process of Sideritis species is lacking.…”

“…It well explains the nature of data set aiming to make arithmetical prediction/forecasting. Compared to conventionally used single‐factor optimization, RSM is more favorable due to the fact that it saves time, sample usage, and space (Lee et al, ).…”

“…As the optimal condition (Lee et al ., ), two grams of Corni fructus powder was extracted with 100 mL of 64% (v/v) aqueous ethanol for 38 min at 78 °C. The remaining steps were the same with the UCGE method.…”

“…Extraction is the most extensively used methodology in the process of natural product purification. Conventional solvent extraction method has been reported for the extraction of iridoid glycosides from Corni fructus (Lee et al, 2012) using volatile and hazardous organic solvent. Ultrasound-assisted extraction is a novel and improved strategy for edible fruits extraction (Zou et al, 2010;Xu & Pan, 2013), which not only increases extraction efficiency but also minimises the consumption of unpleasant petroleum-based solvents (Rostagno et al, 2003;Zuo et al, 2004;Wang et al, 2008).…”
