Mycosphere 8(7), 886-898, Doi 10.5943/mycosphere/8/7/5 AbstractCordycepin (3′-deoxyadenosine), a nucleoside analog, was isolated from Cordyceps militaris, an entomopathogenic fungus, important in Traditional Chinese Medicine. In this study, Cordycepin production by three strains of C. militaris (strains GACP08Y5, GACP08Y1 and GACP0746) in static liquid culture was established using different working volumes and bioreactors. The best cordycepin production of 3005.83 mg/L was obtained by strain GACP08Y5 in 5 L-flasks, containing 2 L medium at day 40, and total cordycepin content reached 6011.66 mg/flask. The utilization ratio of adenine reached 91%. This is the highest report of cordycepin production in a single fermenter. This method provides an effective way for increasing the cordycepin production at a large scale. The strategies used in this study could have a wide application in other fermentation processes.

Cordycepin (COR) has attracted extensive attention for its medical value. Currently, separation and extraction from Cordyceps militaris is the main way to obtain COR, but it is time‐consuming and low efficiency. Heterologous synthesis of COR holds great promise for its industrial production. In this work, five COR synthases of different sources were obtained by homology screening, and were integrated into the genome of BY4741. Fermentation and determination of COR found that EA1 and EA2 from Emericellopsis atlantica were more suitable for COR synthesis. Then, ADO1 gene encoding adenosine kinase was knocked down, ADE4 gene encoding amidophosphoribosyltransferase and truncated cpdBN gene encoding 2′,3′‐cyclic‐nucleotide 2′‐phosphodiesterase/3′‐nucleotidase were overexpressed to enhance the accumulation of COR. Finally, the COR titer reached 725.16 mg L−1 in 5‐L bioreactor. This study lays a foundation for the heterologous synthesis and industrial production of COR, and also provides a reference for heterologous synthesis of other high value‐added chemicals.

Artificial cordyceps mycelium (CM) is a potential substitute for natural cordyceps. In this study, rice bran hydrolysate (RBH), from the residual of the food process, was applied as substrate for submerged fermentation. The active compounds’ constituents in CM were investigated in detail. Results indicated that RBH could not only be used as nutriments and facilitate the submerged fermentation but also showed the ability to promote the metabolism of active ingredients by Hirsutella sinensis HS 1201. Key parameters of the fermentation process were optimized. Under the optimized conditions (pH 6.5, silkworm chrysalis powder 4 g/L, and yeast extract 6 g/L), 15.1 ± 0.6 g/L of the CM was obtained, which was 77.6% higher than the control group that using the conventional synthesis medium. For the constitution of the bioactive compounds, the adenosine content in the CM reached 176 ± 11 mg/100 g, which was increased by 11.4% compared to the control. The concentration of ergosterol in the dry CM was 320 ± 20 mg/100 g, and the cordycepin concentration was 9.66 ± 0.55 mg/100 g. At the same time, lipid yield was 1.2 times higher than that obtained in the control group. The cordyceps polysaccharide content was also increased to 162.3 ± 8.6 mg/g using the RBH‐based medium.
