Most common methods of delivery include the preferred topical (skin), transmucosal (nasal, buccal, sublingual, vaginal, ocular and rectal) and inhalation routes. The conventional dosage forms provide drug release immediately and it causes fluctuation of drug level in blood depending upon dosage form. Therefore to maintain the drug concentration within therapeutically effective range needs novel drug delivery system. In the past few decades, considerable attention has been focused on the development of novel drug delivery system (NDDS). The NDDS should ideally fulfill two prerequisites. Firstly, it should deliver the drug at a rate directed by the needs of the body, over the period of treatment. Secondly, it should channel the active entity to the site of action. In conventional drug delivery systems, there is little or no control over release of the drug and effective concentration at the target site can be achieved by irregular administration of grossly excessive doses. At present, no available drug delivery system behaves ideally, but sincere attempts have been made to achieve them through various novel approaches in drug delivery.

Among the various species of the genus Calendula, C. officinalis is the only one, which is extensively used clinically throughout the world. Pharmacological studies reveal that C. officinalis exhibits antibacterial, antiviral, anti-inflammatory, anti-tumor and antioxidant properties, helps promote the healing of minor burns, scrapes and skin irritations and relieves sunburn and minor cuts. The objective of this study was to optimize conditions for encapsulating Calendula officinalis Linn extract. Thus a novel approach to effectively treat wrinkles, helps promote the healing of minor burns, scrapes and skin irritations and relieves sunburn and minor cuts by delivery of an antioxidant using a special lipid vesicular carrier, the Ethosomal cream was prepared.

A series of new N-(substituted)-5-phenyl-1,3,4-thiadiazol-2-amine derivatives were synthesized under microwave irradiation and evaluated for their anti-inflammatory activity and in-silico (molecular docking studies) to recognize the hypothetical binding motif of the title compounds using VLifeMDS software. The binding mode of the title compounds has been proposed based on the docking studies. They have interesting pharmacophore that display a broad spectrum of biological activity. The 1,3,4-thiadiazole scaffold is an interesting building block that has been used to synthesize a variety of useful bioactive compounds. The present studies widen the scope of the brine shrimp model that may prove quite helpful as a preliminary screen to determine toxic properties. In Brine shrimp lethality bioassay, compounds produced dose dependent cytotoxicity effect to brine shrimp nauplii. Inflammation is a complex process, which is frequently associated with pain and involves occurrences such as the increase of vascular permeability, increase of protein denaturation, and membrane alteration. The pharmacological evaluation of 1,3,4-thiadiazole derivatives revealed that, among all the compounds screened compound code 3c were found to have promising anti-inflammatory activity.
