Optimal composition and position of histidine-containing tags improves biodistribution of 99mTc-labeled DARPin G3

Abstract: Radionuclide molecular imaging of HER2 expression in disseminated cancer enables stratification of patients for HER2-targeted therapies. DARPin G3, a small (14 kDa) engineered scaffold protein, is a promising probe for imaging of HER2. We hypothesized that position (C-or N-terminus) and composition (hexahistidine or (HE) 3) of histidine-containing tags would influence the biodistribution of [ 99m Tc]Tc(CO) 3-labeled DARPin G3. To test the hypothesis, G3 variants containing tags at N-terminus (H 6-G3 and (HE) 3… Show more

“…A new class of targeting molecules, engineered scaffold proteins (ESPs), is promising agents for tumor-targeted delivery of radionuclides [1]. Several types of ESPs, such as affibody molecules [2], ADAPTs [3], and DARPins [4][5][6][7][8][9] demonstrated efficient tumor targeting and high contrast of radionuclide imaging in preclinical studies. DARPins are built of 4-6 repeat modules of 33 amino acids and have a molecular weight of 14-18 kDa [10].…”

“…DARPins have both of these properties and can be successfully utilized for the radioactive imaging of tumors. For example, HER2-specific DARPins G3 and 9_29 were used for obtaining conjugates with the desired pharmacokinetics and reduced accumulation in the liver [47][48][49]. As for cancer therapy, DARPins can be used both for the delivery of toxic modules and for the inhibition of cell signaling pathways thanks to the specific binding of membrane receptors.…”

“…It was shown that the IL-37-IL-1R8-IL-18Rα complex is assembled on the cell surface, and that its presence is necessary for further triggering of an anti-inflammatory response [46]. The IL-1R8 receptor consists of only one extracellular Ig-like domain with a long "tail" immersed in the cytoplasm and containing a mutant TIR domain [47]. The involvement of IL-1R8 in the anti-inflammatory action of IL-37 has been demonstrated in experiments on mice defective in the gene for this chain.…”
