Optical Studies of Nanodiamond-Tissue Interaction: Skin Penetration and Localization

Abstract: In this work, several optical-spectroscopic methods have been used to visualize and investigate the penetration of diamond nanoparticles (NPs) of various sizes (3–150 nm), surface structures and fluorescence properties into the animal skin in vitro. Murine skin samples have been treated with nanodiamond (ND) water suspensions and studied using optical coherence tomography (OCT), confocal and two-photon fluorescence microscopy and fluorescence lifetime imaging (FLIM). An analysis of the optical properties of th… Show more

“…Such aggregation can prevent transdermal activity, as NDs with a diameter larger than 100 nm will not penetrate to the deep skin layers. Rather, they will be localized mostly in appendages that are open to the skin, mainly hair follicles, but also in sweat gland pores and the sebaceous gland . Those appendages allow the NDs to cross the stratum corneum, which is in coherence with experiments done on other NPs .…”

“…Test TiO 2 NP solutions were prepared immediately before use through dilution of the stock solutions with distilled deionized water and sonication (50 W/L, 40 kHz) for 20 min. In our studies, the particle size distributions and z-potential were estimated using the dynamic light scattering method (DLS) with a Zetasizer Nano ZS (Malvern Instruments, Malvern, UK) [36,37]. The averaged z-potential of the used TiO 2 NPs in solution was 22.08 ± 0.32 mV, with pH value around 5.35-5.45.…”
