A new light-chain marker has been detected in normal mouse serum immunoglobulin light chains by gel isoelectric focusing. The marker (Ef2) involves the presence of two major and several minor bands in the normal light-chain IF profiles. Strains expressing the marker IF bands are designated Igk-Ef2a, whereas those lacking the bands are Igk-Ef2b. The majority of inbred strains are Igk-Ef2a. Strains found to be Igk-Ef2b are NZB/BlNJ, BDP/J, C58/J, I/LnJ, CE/J, and P/J. The strain distribution of the alleles differs from the distribution of alleles at the Ly-2 and Ly-3 loci, suggesting the new marker may represent a separate locus. Genetic studies have shown that Igk-Ef2 locus is closely linked to Igk-Ef1 and Hd loci on Chromosome 6, indicating that it is also closely linked to Ly-3. The relative importance of the bands controlled by the Igk-Ef2 locus suggests that the entire normal light-chain pool could be controlled by as few as 100 such loci.

The genetic control of the variable (V) 1 regions of mouse antibodies is being analysed at several levels. The direct chemical analysis of V-region genes is underway [87]; antibody V-genes have been isolated from the D N A of differentiated and undifferentiated mouse cells and are now being sequenced. These studies already provide support for the conclusions based on analysis of amino acid sequences of V-regions and the genetic analysis of V-region genes using markers such as idiotype. In this chapter, we will concentrate on what has been learned from the latter kinds of studies such as the approximate size of VL and V H loci, the organization of VH genes and the relationship of V-region and C-region loci. Our purpose will be to bring to date the information on mouse V regions from a variety of sources using as a basis several recent reviews of this field by Eichmann [27], M~ikel~i and Karjalainen 1-59], Potter [74] and the summaries of the H o m o g e n e o u s Immunoglobulin Workshops [90].V-region genetic studies in the mouse owe their rapid development to two systems: the first of these is the mouse plasmacytoma developed by Michael Potter and co-workers. As Melvin Cohn points out, "The mouse system bears the stamp of Michael Potter who systematically developed the methods for provoking and characterizing mouse myelomas and who established a collection of cell lines which are to this field what the T-phages were to molecular biology" [24]. The difficulty to comprehend the immune response resides both in the complexity of antigens, in terms of linear structure and arrangement in space, as the eliciting signal and the complex molecular and cellular interactions that result in the induction and the expression of antibodies. In addition, every antibody response is the sum of many clonal responses, specific and cross-reactive. Hence, any study dealing with the expression of antibodies calls in the last analysis for means to separate the antibody forming clones and their products, the monoclonal antibodies (9). The analysis of myeloma proteins (37) as an alternative for antibodies with predetermined specificities can only be regarded as a substitute for the issue to be addressed -these substrates are the final products that rarely have known antigen specificity and cannot tell the story leading to their expression -until a unifying model will become available to resolve the structure-function relationship of antibodies, the connections between specific antibodies and correlating T cell receptor specificity, and the genetics of the immune response and of the antibody variable regions.A number of different antigen systems and of antibody models have been used to tackle these problems (18). Undoubtedly, the streptococcal group polysaccharide antigens have played a significant role in this line of research, since antibody responses of restricted heterogeneity to these bacterial cell wall antigens were reported (28,35), and could later be elicited at will and in a predictable fashion both in selectively bred rabbits and in certain inbred strains of mice (5,8, IS,29).
