Oligodendrocytes as glucocorticoids target cells: functional analysis of the glycerol phosphate dehydrogenase gene

Abstract: Previous research has established that the development and function of oligodendrocytes are influenced by glucocorticoids. The enzyme glycerol phosphate dehydrogenase (E.C.1.1.1.8) has been used as a model to study glucocorticoid regulation of gene expression in oligodendrocytes and the C6 glial cell line. In the rat brain this enzyme is exclusively localized to oligodendrocytes. The sequence of the 5' flanking region for the rat gene encoding Glycerol Phosphate Dehydrogenase (GPDH; EC 1.1.1.8) was determined.… Show more

“…We had reported activation of NFjB in response to demyelination and GFs-treated remyelination [1]. Among the cellular effectors involved in the neutralization of oxidative species, cytosolic glycerol phosphate dehydrogenase (cGPDH) is expressed in OLs and have been described for its role in stress [20][21][22][23][24]. A 50% increase in the enzymatic activity of cGPDH occurred in a three-week treatment of mice with cuprizone (Fig 9a).…”

“…We base this conclusion on the accumulation of Formation of glycerol from glucose requires, in addition to the enzymes of glycolysis from hexokinase to aldolase, the presence of glycerol-3-phosphate dehydrogenase, which can convert dihydroxyacetone phosphate into glycerol-3-phosphate, and a phosphatase that can dephosphorylate glycerol-3-phosphate to glycerol. Cytosolic glycerol-3-phosphate dehydrogenase is expressed by oligodendrocytes (Leveille et al 1980;Cheng and de Vellis 2000), and mitochondrial glycerol-3-phosphate dehydrogenase has been shown to be expressed by neurons and astrocytes (Nguyen et al 2003).…”

“…Conversion of dihydroxyacetone phosphate into glycerol-3-phosphate has different biochemical consequences depending on whether the reaction is catalyzed by the cytosolic, NAD + -dependent, or the mitochondrial, FAD-dependent, glycerol-3-phosphate dehydrogenase. The cytosolic reaction would regenerate NAD + in the cytosol of oligodendrocytes, as this enzyme is oligodendroglial, not neuronal or astrocytic (Leveille et al 1980;Cheng and de Vellis 2000). In contrast, the mitochondrial reaction would oxidize mitochondrial FADH 2 to FAD.…”
