“…Another interesting aspect of the anti-cytotoxic action of OleA, in the contest of amyloydosis, was reported by Leri et al [130]. Using another amyloidogenic protein, a variant of human β2-microglobulin (β 2 m) [131][132][133] a 99 residue-long human protein belonging to the major histocompatibility complex class I (MHC I) associated with a familial form of systemic amyloidosis, they showed that it exhibited an enhanced amyloidogenic tendency to aggregate in vitro with respect to the wild-type protein.…”

“…Therefore, in an additional set of experiments, we investigated the ability of the Aβ 42 aggregates grown in the presence of ARE (100 µg/mL) to interact with the cell membrane of exposed cells with respect to the aggregates grown in the absence of ARE. To this purpose, we performed confocal microscopy experiments using a polyclonal antibody raised against recombinant Aβ 42 and Alexa 488-conjugated CTX-B, a probe specific for the monosialotetrahexosylganglioside 1 (GM1), a common lipid raft marker widely reported as a key interaction site for amyloids [22, 25]. Aβ 42 fibrils interaction with GM1 was evaluated by FRET analysis (Fig.…”

“…Animal studies showed a higher level of autophagy, while cell culture experiments suggest that probably oleuropein aglycon regulates mTOR protein [87]. Recent studies confirmed the activity of oleuropein aglycon against amyloid aggregation by modification of the conformational and biophysical properties of amyloid fibrils and of the cell bilayer surface properties at the same time, remodeling the aggregation and helping the association between the protein and the membrane [88]. In AD situation, it was observed various Aβ peptides in amyloid plaques were observed, characterized by N-terminal truncation.…”
