Nowadays, Huanglongbing (HLB) disease, associated with Candidatus Liberibacter asiaticus (CLas), seriously affects citriculture worldwide, and no cure is currently available. Transcriptomic analysis of host-pathogen interaction is the first step to understand the molecular landscape of a disease. Previous works have reported the transcriptome profiling in response to HLB in different susceptible citrus species; however, similar studies in tolerant citrus species, including Mexican lime, are limited. In this work, we have obtained an RNA-seq-based differential expression profile of Mexican lime plants challenged against CLas infection, at both asymptomatic and symptomatic stages. Typical HLB-responsive differentially expressed genes (DEGs) are involved in photosynthesis, secondary metabolism, and phytohormone homeostasis. Enrichment of DEGs associated with biotic response showed that genes related to cell wall, secondary metabolism, transcription factors, signaling, and redox reactions could play a role in the tolerance of Mexican lime against CLas infection. Interestingly, despite some concordance observed between transcriptional responses of different tolerant citrus species, a subset of DEGs appeared to be species-specific. Our data highlights the importance of studying the host response during HLB disease using as model tolerant citrus species, in order to design new and opportune diagnostic and management methods.

El huanglongbing (HLB) es la enfermedad más devastadora de los cítricos a nivel mundial y está presente en el estado de Colima desde 2010, zona productora de limón mexicano (Citrus aurantifolia). Esta enfermedad causa un desequilibrio en el reparto de carbohidratos e inhibe la fotosíntesis. También afecta la concentración de nutrientes en hoja. El objetivo de este trabajo fue determinar los cambios fisiológicos en árboles en producción de limón mexicano infectados con HLB. El estudio se desarrolló en Colima en 2017. Se determinó la concentración de almidón, sacarosa y glucosa en nervadura central y lámina de hojas de diferentes edades (120 y 30 días) y almidón en raíces. La concentración de clorofilas por método analítico e índice SPAD en hojas con diferentes niveles de síntomas de HLB y análisis foliares de nutrientes minerales. Las hojas con HLB tuvieron mayor concentración de almidón en la lámina de 120 días. La sacarosa se concentró en las nervaduras de hojas de 120 días. Mientras que la glucosa se incrementó en las nervaduras de 30 y 120 días. Las raíces de árboles infectados con HLB mostraron 40% menor concentración de almidón comparados con árboles sanos. Disminución en el contenido de clorofilas e índice SPAD en función del grado de afectación. Así como menor concentración de K, Ca, Mg, Cu, Fe, Zn y B. El conocer los cambios que ocasiona el HLB en limón mexicano permitirá desarrollar prácticas que disminuyan su impacto.

Huanglongbing (HLB), also known as citrus greening, is a destructive disease of citrus; it is considered a newly emerging disease which has spread to the Middle East and North Africa (MENA). In Iran, the disease was first found in 2009. In this study, two hypervariable prophage and phage‐related loci, bacteriophage repressor protein C1 (CLIBASIA_ 01645 locus) and prophage terminase gene (CLIBASIA_05610 locus), were used to determine the diversity and characterization of Candidatus Liberibacter asiaticus (CLas) strains associated with HLB samples. Analyses of the CLIBASIA_01645 locus, characteristic of variable tandem repeat numbers (VTRNs), revealed the homogeneity of Iranian CLas isolates: However, this result showed two distinct genotypes (TRN < 10 and TRN > 10) of CLas in Iran. This is the first report documenting the presence of two differentially distributed genotypes of CLas in Iran. Sequence analysis of prophage terminase revealed the presence of two putative prophages (prophage I and prophage II) in the genome of CLas isolates of Iran. Frequency analysis of these two prophages by specific loci revealed the association between prophages populations, the development HLB symptoms and CLas genotypes and their interactions with another obligate symbiontic, HLB phytoplasma.
