Antibiotic pollutions in the aquatic environment have attracted widespread attention due to their ubiquitous distribution and antibacterial properties. The occurrence, distribution, and ecological risk assessment of 17 common antibiotics in this study were preformed in a vital drinking water source represented as a river-reservoir system in South China. In general, 15 antibiotics were detected at least once in the watershed, with the total concentrations of antibiotics in the water samples ranging from 193.6 to 863.3 ng/L and 115.1 to 278.2 µg/kg in the sediment samples. For the water samples, higher rain runoff may contribute to the levels of total concentration in the river system, while perennial anthropic activity associated with the usage pattern of antibiotics may be an important factor determining similar sources and release mechanisms of antibiotics in the riparian environment. Meanwhile, the reservoir system could act as a stable reactor to influence the level and composition of antibiotics exported from the river system. For the sediment samples, hydrological factor in the reservoir may influence the antibiotic distributions along with seasonal variation. Ecological risk assessment revealed that tetracycline and ciprofloxacin could pose high risks in the aquatic environment. Taken together, further investigations should be performed to elaborate the environmental behaviors of antibiotics in the river-reservoir system, especially in drinking water sources.

Bacterial resistance to carbapenem, which is mainly due to the successful dissemination of carbapenemase-encoding genes, has become a major health problem. Few studies have aimed to characterize the level of resistance in the environment, notably in hospital wastewater, which is a likely hotspot for exchange of antibiotic resistance genes. In this work, we looked for the presence of imipenem-resistant bacteria and imipenem in the effluent of the teaching hospital of Clermont-Ferrand, France. Selective growth of bacteria from 14-day old biofilms formed in the pipe sewer showed that 22.1% of the isolates were imipenemresistant and identified as Aeromonas (n=23), Pseudomonas (n=10), Stenotrophomonas (n=4) and Acinetobacter (n=1). Fifteen of these strains harbored acquired carbapenemase-encoding genes bla VIM (n=11), bla OXA-48 (n=2), bla GES (n=1), bla NDM (n=1). All isolates also harbored associated resistances to aminoglycosides, fluoroquinolones and/or tetracyclin. S1-nuclease pulsed-field gel electrophoresis analysis of eight selected isolates showed that four of them harbored one to two plasmids of molecular weight between 48.5Kb and 194Kb. In vitro transformation assays evidenced the presence of bla VIM and bla NDM on plasmids with the bla VIM harboring 80 Kb plasmid having conjugative capacity. The predicted environmental concentration of imipenem in the hospital effluent was 3.16 µg/L, suggesting that biofilm bacteria are subjected to sub-MICs of imipenem within the effluent. However, no imipenem molecule was detected in the hospital effluent, probably owing to its instability: in vitro assays indicated that imipenem's biological activity was no longer detectable after 45h of storage. However, the predictive value of the hazard quotient relative to the development of resistance was greater than 1.0 (HQr=28.9 ± 1.9), which indicates a possible risk. The presence of carbapenemase-encoding genes in hospital effluent biofilm strains and their ability to transfer are therefore a potential hazard that should not be neglected and points to the need for monitoring antibiotic resistance in hospital wastewater.

Abstract. The need for effective chemical water monitoring is evident. Indeed the overall management and decision making system driven by the Water Framework Directive (WFD) and other European water policies is strongly dependent on monitoring data. Because the overall decision making process relies, most of the time, on acquired data, it puts considerable pressures on the display of high quality chemical environmental measurements. However, performing fit for purpose measurements implies i) the demonstration of their metrological traceability ii), the evidence of their achievement thanks to accurate and sensitive analytical methods and iii) their statement with a realistic estimate of expanded uncertainty are thoroughly addressed. Concern regarding the presence of potentially active substances in surface waters was brought up by the end of the 90s. Intensive monitoring programs have revealed a chronic state of contamination of water systems by numerous molecules; pharmaceuticals have been highlighted as one of first concern for French aquatic system. This work demonstrates the consequences that (i) the sampling strategy and (ii) methodological and (iii) metrological laboratory choices may have on the measurements' result. It reemphasizes that the main contribution to the uncertainty of measurement is linked to sampling uncertainty
