Adipose tissue-derived stem cells (ADSCs) are mesenchymal cells with the capacity for self-renewal and multipotential differentiation. This multipotentiality allows them to become adipocytes, chondrocytes, myocytes, osteoblasts and neurocytes among other cell lineages. Stem cells and, in particular, adipose tissue-derived cells, play a key role in reconstructive or tissue engineering medicine as they have already proven effective in developing new treatments. The purpose of this work is to review the applications of ADSCs in various areas of regenerative medicine, as well as some of the risks associated with treatment with ADSCs in neoplastic disease.

ResumenObjetivo. Comparar viabilidad, crecimiento en cultivo, fenotipo y diferenciación a linaje osteogénico de ADAS (Adipose-Derived Adult Stem Cells) pre y post congelamiento. Método. Se obtuvieron muestras de TA de individuos sanos que fueron sometidos a liposucción, con posterior digestión enzimática con Colagenasa I. La fracción vascular estromal (FVE) obtenida fue dividida equitativamente en dos fracciones una de las cuales fue criopreservada y almacenada a -196°C y la otra cultivada hasta pase uno antes de ser criopreservada. Ambas fracciones fueron criopreservadas en cámara de congelamiento automatizada utilizando dos soluciones de congelamiento: (A) Dimetilsulfóxido (DMSO)/Dextrano40/albúmina sérica humana (HSA) y (B) DMSO/HESSICO/HSA, mantenidas 3 meses a -196°C y descongeladas con buffer fosfato salino (PBS)/HSA/HESSICO. Posterior a su descongelamiento fueron sometidas a cultivo, recuento, viabilidad celular, fenotipificación y potencial osteogénico. Resultados. Las células congeladas con la solución crioprotectora DMSO/Dextrano40/ HSA y con un cultivo previo al congelamiento, presentaban porcentajes de viabilidad más altos y alcanzaban confluencia del 80% en cultivo en menos tiempo. Las muestras expandidas no presentaron diferencias en el fenotipo por citometría de flujo y mostraron diferenciación a linaje osteogénico.Palabras clave: células madre mesenquimales, tejido adiposo, criopreservación.
AbstractObjective. Comparing viability, growth in culture, phenotype and osteogenic lineage differentiation of Adipose-Derived Adult Stem Cells (ADAS) pre and post freezing. Method. Samples were obtained from adipose tissue of healthy individuals who underwent liposuction, with later enzymatic digestion with collagenase I. The stromal vascular fraction (SVF) obtained was divided equally into two fractions one of which was cryopreserved and stored at -196 °C and the other one cultivated to passage 1 (P1) before being cryopreserved. Both fractions were cryopreserved in an automated freezing chamber using two freezing solutions: (A) DMSO/Dextran40/HSA and (B) DMSO/HESSICO/HSA kept, both were kept 3 months at -196°C and thawed in PBS/HSA/ HESSICO. After thawing, both were subjected to cultivation, recount, cell viability, phenotyping and osteogenic potential. Results. Cells that were frozen with cryoprotectant solution DMSO/ Dextran40/HSA and a pre-freezing culture exhibited higher viability percentages and reached 80% culture confluence in less time. Expanded samples showed no differences in the phenotype by flow cytometry and showed differentiation to osteogenic lineage.

ResumenEl objetivo de la presente revisión sistemática es determinar si los lipoinjertos enriquecidos con células madre derivadas del tejido adiposo son superiores a los injertos de tejido graso convencionales en el tratamiento de la lipoatrofia facial.Realizamos una búsqueda sistemática de publicaciones en las bases de datos: MEDLINE, SCOPUS, OVID SP, SCIELO y EMBASE y posteriormente, una valoración basada en criterios actuales de medicina basada en la evidencia de las publicaciones encontradas por dos revisores independientes; las discrepancias en el análisis se resolvieron por acuerdo. Los criterios de inclusión fueron: ensayos clínicos aleatorizados, series clínicas y reportes de casos en humanos, excluyendo los ensayos en animales.Encontramos 6 publicaciones que cumplían con los criterios previamente mencionados. Entre éstas se incluyeron 2 ensayos clínicos aleatorizados controlados que recogían la superioridad de los lipoinjertos enriquecidos con células madre derivadas del tejido adiposo en el tratamiento de la lipoatrofia facial en términos de resistencia a la reabsorción (preservación del volumen) y menor necesidad de nuevos lipoinjertos, en comparación con los lipoinjertos convencionales.Concluimos diciendo que existen publicaciones con buenos niveles de evidencia que sugieren que los lipoinjertos enriquecidos con células madre derivadas del tejido adiposo son superiores a los lipoinjertos convencionales en el tratamiento de la lipoatrofia facial. AbstractThe objective of this systematic review is to determine if the lipografts enriched with stem cells derived from adipose tissue are superior to the conventional fatty tissue grafts in the treatment of facial lipoatrophy.A systematic search was performed for publications in the databases: MEDLINE, SCOPUS, OVID SP, SCIELO and EMBASE. An evaluation was subsequently conducted based on current criteria of evidence-based medicine of the publications found by two independent reviewers and discrepancies in the analysis were resolved by agreement. The inclusion criteria were: randomized clinical trials, clinical series and case reports in humans; the reports in animals were excluded.We found 6 publications that meet the criteria previously mentioned. Between these, 2 randomized controlled clinical trials reported the superiority of the lipografts enriched with stem cells derived from adipose tissue in the treatment of facial lipoatrophy in terms of resistance to the reabsorption (volume preservation) and less need for new lipografts when compared with the conventional lipografts.We conclude that there are publications with good levels of evidence to suggest that the lipografts enriched wi...
