Effect of chemical and heat therapy on virus concentrations in in vitro potato plantlets. Can. J. Bot. 68: 1515-1521.Potato (Solanurn tuberosum) plantlets were established in vitro to evaluate the effectiveness of chemicals and heat therapy for antiviral activity. Ribavirin was shown to be the most active chemical. Further studies were performed in which plantlets established from nodal cuttings were exposed to an alternating 4-h cycle of 35-31°C and ribavirin (20 mg/L) therapy for 4 weeks, and then tested quantitatively for virus titer by the enzyme-linked immunosorbent assay. Plantlets were further propagated via nodal cuttings at room temperature without exposure to ribavirin. Virus assays were performed and virus-free plantlets were grown out as mature plants for a final virus assay. Ribavirin alone or in combination with heat therapy was effective in reducing potato virus M, S, and X titers (between 10-and 60-fold). Virus-free plants were obtained from both treatments. For potato viruses Y and leafroll, about a fourfold reduction in titer was observed after the ribavirin-heat treatments. When multiple viruses were present, quantitative (10-to 20-fold) reductions in potato viruses M, S, X, and leafroll were observed and resulted in plants free of potato viruses M, S, and X. This protocol enables the investigator to quantitatively monitor the impact of a single variable on the virus-host interaction. Lilium brownii var. colchesteri has unique and ornamental floral characteristics in graceful harmony with flower and anther colors, flower shape, elegant fragrance, and flower color change from yellowish cream to white during anthesis. There are, however, few accessions conserved in Japan up to the present, and they often show abnormally shaped flowers and mosaic leaves seemingly due to virus infection. Virus-free bulblets were established by combining meristematic tip culture and chemotherapy. At initial diagnosis with RT-PCR, Cucumber mosaic virus (CMV), Lily mottle virus (LMoV) and Lily symptomless virus (LSV) were detected from leaf tissues of the mother plants. All regenerated bulblets obtained by meristematic tip culture of the mother plants were still infected by at least one of the viruses. The meristematic tip of the bulblets infected with either LSV or LMoV was selected for subsequent culture with 2,4-dioxohexahydro-1,3,5-triazine (DHT), an antiviral chemical. LSV was eliminated successfully in mericlones from bulblets with LSV, whereas LMoV was not from those with LMoV. The virusfree bulblets were transferred to new medium without DHT, and multiplied by in vitro scaling. They were then acclimated in a phytotron glass room at 20°C. The plants were confirmed to be virus-free after 18-months' acclimation. It was concluded that the combination of meristem tip culture and chemotherapy is practical for producing virus-free plants of L. brownii var. colchesteri.

Viruses are microorganisms that infect all kinds of living organisms including plants, and cause remarkable lose in crop production. Although pesticides showed that they can protect plants from pest infections, there are no effective substances that can be used as potent virucides. Therefore, there is a continuous demand to produce chemicals in order to stop and cure viral infections in plants. However, toxicity and carcinogenicity issues were always attributed to chemical pesticides. Screening of natural products shined in the dark to find new safe virucides. The philosophy of selecting those plants is oriented towards plants that can protect themselves from viral infections. Plants have been reported as virus inhibitors and are able to prevent infection of viruses by inducing systemic resistance in noninfested parts of the plants, such as Boerhaavia diffusa, Clerodendrum aculeatum. Other plants defend themselves against virus infections; these plants contain ribosome inactivating proteins [RIPs], such as Phytolacca Americana, Mirabilis jalapa, Dianthus caryophullus. The methods used for extraction, separation, identification of those antiviral compounds are documented and discussed in this review.
