Höller and Breucr: The effect of fluorocarbon FC 43 on the metabolism of steroids during perfusion of the isolated rat liver 3 Summary: The influence of fluorocarbon FC 43 on the metabolism of oestrogens in the isolated perfused rat liver was investigated. In comparative once-through pcrfusions with FC 43 emulsion or albumin solution as perfusion medium, the clearance rates of oestrone and its metabolites were determined. In pcrfusions with FC 43, the clearance of oestrone was lower, and the metabolites formed in the liver were more concentrated in the outflowing medium than in perfusions without fluorocarbon. This can be explained by the high affinity, even of conjugated oestrogens, to FC 43, which is established by equilibrium dialysis and partition coefficient. The results presented here show that the fluorocarbon has a strong influence of its own on the metabolism of steroids in the isolated perfused liver. Therefore, this solvent should be avoided as medium when the metabolism of steroids is studied in perfusion experiments. Einfluß von Fluorcarbon FC 43, auf den Stoffwechsel von Steroiden während Perfusion der isolierten RattenleberZusammenfassung: Der Einfluß von Fluorcarbon FC 43 auf den Stoffwechsel von Steroiden in der isoliert perfundierten Rattenleber wurde untersucht. Zu diesem Zweck wurde eine Emulsion mit 150 ml/l FC 43 oder eine Pufferlösung mit 30 g/l Albumin nicht-zyklisch durch Rattenlebern pcrfundiert. Beide Medien enthielten gleiche Konzentrationen von [4-14 C]Östron. Das ausfließende Medium wurde fraktioniert aufgefangen und analysiert. Es zeigte sich, daß die Clearance von Östron bei Perfusionen mit FC 43-haltigem Medium deutlich niedriger ist als in Versuchen mit fluorcarbonfreiem Medium; gleichzeitig werden die in der Leber entstehenden Metaboliten auf Grund ihrer Affinität zu FC 43 schneller an das Medium abgegeben. Die relativ große Affinität wurde durch Gleichgewichtsdialyse und Untersuchungen über den Verteilungskoeffizienten nachgewiesen. Die hier vorgelegten Ergebnisse führen zu dem Schluß, daß ein Medium, welches Fluorcarbon FC 43 enthält, zum Studium des Stoffwechsels von Steroiden in der isoliert perfundierten Rattenleber nicht geeignet ist.

This volume is an update of the Solubility Data Series Volume 7 published in 1981 on oxygen and ozone. For this volume, the literature was covered from 1981 to 2009 with some papers from 2010. Some earlier papers missed in the 1981 volume are included. Interest in the solubility of oxygen and of ozone continues to be high. The solubility of oxygen in water at low pressure seems well established, but more work is needed on the solubility of oxygen in water at high pressure and high temperature. There is now a wealth of salt-effect data on the solubility in aqueous salt solutions, but some glaring discrepancies still exist. Little has been done on salt effects at high pressure and high temperature. The solubility data of oxygen in hydrocarbons seem sparse. The solubility of oxygen in alcohols through C 5 seems well established. Between the 1981 volume and this volume, there are solubility data in over 70 fluorocarbons and fluorocarbon compounds with O, H, S, and N. A new area is the solubility of oxygen in ionic liquids, which are becoming increasingly important as replacement solvents for volatile organic solvents. Perfluorochemical Perfusion of the Isolated Guinea Pig Heart

We studied the influence of a perfluorochemical (PFC) emulsion on the ultrastructure and function of the isolated perfused guinea pig heart compared to a Krebs-Henseleit solution. The PFC perfusion enhanced the force of contraction and reduced the coronary flow rate, but had no influence on the frequency and the oxygen consumption. The positive inotropic action of K-strophanthin and isoproterenol was slightly strengthened, whereas the β-adrenergic antagonism by propranolol and the vasodilatation by glycerol trinitrate remained unchanged. The positive chronotropic action of isoproterenol was reduced during PFC perfusion. No histological differences depending on the perfusion medium were observed. It is concluded that PFC perfusion improves the functional state of the Langendorff preparation.
