Nucleotide Sequence of Narcissus Mosaic Virus RNA

Abstract: SUMMARYThe nucleotide sequence of the genomic RNA of narcissus mosaic virus (N MV) was deduced from a set of cDNA clones and by direct sequencing of RNA. The genome, with a length of 6955 nucleotides [excluding the 3'poly(A) tail], contains six open reading frames (ORFs) with the capacity to code for polypeptides of more than 1 OK, with Mr values of 186284, 25845, 13998, 11059, 26097 and 10519. The first five of these putative proteins show considerable homology to similar proteins encoded by the RNAs of potat… Show more

“…The extreme Y-terminal sequence of FMV RNA, GAAAACUCUUCC, is very similar to the terminal sequence of other potexviral RNAs (Zuidema et al, 1989). The complete nucleotide sequence of FMV RNA is presented in Fig.…”

“…The remainder of ORF2 sequences from all other potexviruses are less related to each other than are ORF1 and ORF3 (IlK protein) sequences (Table 3; see also Zuidema et al, 1989). FMV ORF 3 and ORF 4 are related to those of other potexviruses in that they contain stretches of hydrophobic residues predicted to form transmembrane helices (Morozov et al, 1987;Forster et al, 1988;Skryabin et aL, 1988b) bordered by charged residues.…”

“…This produced clone Nco4B which contained a net addition of two amino acid residues within the conserved carboxy-terminal region of the polymerase gene (Zuidema et al, 1989). The second mutant, PL6, was constructed at the unique EcoRI site (position 1894) within the non-homologous region of the polymerase gene (see Fig.…”

“…2A). The conserved hexanuleotide, ACCUAA, involved in accumulation of all known potexviral RNAs [52][53][54][55][56] was found in the apical loop of SLC. A putative polyadenylation signal, AAUAAA, important for the infectivity of BaMV and White clover mosaic virus (WClMV) 53,57 was identified downstream of SLC.…”
