Nucleic acid-specific suppressor T cells.

Abstract: The concept of using cell-bound antigens as tolerogen was applied to nucleic acid. Nucleoside was linked directv to s leen cell suspensions. Intravenous administration of nucleoside coupled to isogeneic spleen cells into mice generated suppressor cells that diminished the formation of antibody-forming cells either to a T-dependent antigen in vivo or to a T-independent antigen in vitro. Suppressor cells were nucleoside specific, but the specificity of immune suppression seems to be somewhat broader than that of… Show more

“…More recently, suppressor T cells specific for nucleosides have been elicited in vivo [8,40]. The latter finding might be of importance not only for therapy, but also for exploring the role of suppressor T cells specific to nucleic acid in the pathogenesis of systemic lupus erythematosus.…”

“…The dichotomy between these two phenomena is illustrated by several observations dealing with both the cellular mechanism and the specificity of unresponsiveness [6]. For example, whereas guanosine linked to IgG induces guanosine specific tolerance only, guanosine linked to cells suppresses antibodies not only to guanosine, but also to cytosine and adenosine [8,27]. Suppression affects both the expression of cellular (T) and humoral (B) immunity and is mediated by specific suppressor T cells [15,33].…”

“…It is well known that immunological unresponsive ness is induced by the transfer of syngeneic lymphoid cells modified chemically with haptens [1], nucleic ac ids [2], and proteins [3]. With these antigens, suppres sor T cells [4], as well as killer cells [5], are generated in mice.…”

“…It is well known that the immunologic unrespon siveness is induced by the transfer of syngeneic lym phoid cells modified chemically with haptens [1][2][3], nucleic acids [4] and proteins [5], With these antigens, suppressor T cells [1][2][3][4][5], as well as killer cells [6], are produced in mice. However, the chemical modifica tion of lymphoid cells by these substances would not normally occur in vivo except in special circum stances when animals are exposed to chemicals such as DNFB ( I-fiuoro-2,4 dinitrobenzene) or TNBS (2,4,6-trinitrobenzene sulfonic acid), and so on.…”
