Abstract: Our results show that KCS promotes chromatin remodeling in epithelial cells and lymphocytes on the conjunctival surface of dogs. The changes described in this study are different from those reported for conjunctival cell nuclei of human KCS patients.

One hundred years ago, Robert Feulgen published a landmark paper in which he described the first method to stain DNA in cells and tissues. Although a century has passed since the discovery by Feulgen and Rossenbeck, the chemical reaction still exerts an important influence in current histochemical studies. Its contribution in diverse fields, spanning from biomedicine to plant biology, has paved the way for the most significant studies that constitute our current knowledge. The possibility to specifically explore the DNA in cell nuclei while quantifying its content makes it a contemporary and timeless method. Indeed, many histocytochemical studies following the 1924 paper have led to a deep understanding of genome organization in general as well as several specific mechanisms (e.g. DNA duplication or tumour pathology) that, nowadays, constitute some of the most fundamental pillars in biological investigations. In this review, we discuss the chemistry and application of the Feulgen reaction to both light and electron microscopy.

ObjectiveTo evaluate a grading scheme for conjunctival staining patterns with lissamine green ocular dye in the diagnosis of tear film deficiencies in dogs.ProceduresClient‐owned and research colony dogs were enrolled in a prospective study between February and October 2018 in which slit‐lamp biomicroscopy, Schirmer tear test (STT), tear film breakup time (TFBUT), conjunctival lissamine green staining (LGS), and intraocular pressure (IOP) measurement were performed in both eyes of all dogs. Lissamine green staining of the temporal bulbar conjunctiva was graded from 0‐3, with a higher grade corresponding to an increased stain intensity.ResultsFifty‐four dogs (107 eyes), comprising 31 males and 23 females with a mean age of 5.0 ± 3.9 years (range 0.5‐14.3), were enrolled in the study. STT was <15 mm/min in 21 eyes and ≥15 mm/min in 86 eyes. Lissamine green staining grade for eyes with a STT of <15 mm/min (2.0 ± 0.9) was significantly higher than for eyes with a STT ≥15 mm/min (0.2 ± 0.7) (P < .001). TFBUT for eyes with a STT <15 mm/min (6.5 ± 4.4 seconds) was significantly shorter than for eyes with a STT ≥ 15 mm/min (16.1 ± 3.6 seconds) (P < .001). As LGS grade increased, both STT (P < .001) and TFBUT (P < .001) significantly decreased.ConclusionsA higher LGS grade was significantly associated with a lower STT and more rapid TFBUT in dogs. Lissamine green ocular dye can be considered as an adjunctive diagnostic test when evaluating tear film deficiency in dogs.

Dry-eye syndrome in Shih Tzu dog: Ocular surface clinical and morphological aspects in the different stages of keratoconjunctivitis sicca

This study analyzed clinical and cyto-histomorphological parameters of the ocular surface of Shih Tzu dogs, non-carriers and carriers of quantitative keratoconjunctivitis sicca (KCS) at different stages. Thirty-five eyes from 23 male and female Shih Tzu dogs between two and eight years were evaluated in four groups: control group (CG - without KCS), mild KCS group (KCS1), moderate KCS group (KCS2), and severe KCS group (KCS3). Most clinical variables among KCS carrier groups worsened at the more advanced stages of the disease, with a negative correlation between the Schirmer tear test (STT-1) and tear film break-up time (TBUT). Squamous metaplasia, lymphoplasmacytic inflammatory infiltrate, and decrease in conjunctival goblet cells on histopathological examination comprised disease severity parameters. Quantitative KCS non-carried Shih Tzu dogs have qualitative and tear film distribution changes. The cytomorphological exam is limited to evaluating the inflammatory infiltrate and quantifying conjunctival goblet cells. However, intermediate epithelial cells were higher in healthy eyes compared to eyes with KCS in Shih Tzu dogs. Also, moderate and severe KCS carrier Shih Tzu dogs have qualitative dysfunction of the tear film. KCS in Shih Tzu dogs is common and chronic and may be responsible for the loss of vision of these animals. Early identification of the disease and routine evaluation can improve these dogs’ quality of life and ocular health.
