NRF2 Blockade Suppresses Colon Tumor Angiogenesis by Inhibiting Hypoxia-Induced Activation of HIF-1α

Abstract: Transcription factor NRF2 is an important modifier of cellular responses to oxidative stress. Although its cytoprotective effects are firmly established, recent evidence suggesting important roles in cancer pathobiology has yet to be mechanistically developed. In the current study, we investigated the role of NRF2 in colon tumor angiogenesis. Stable RNAi-mediated knockdown of NRF2 in human colon cancer cells suppressed tumor growth in mouse xenograft settings with a concomitant reduction in blood vessel format… Show more

“…The regulation of HIF-1α by Nrf2 has been investigated in tumor angiogenesis. Suppression of Nrf2 inhibited HIF-1α and resulted in a reduction of VEGF and blood vessel formation in human glioma cells (17) and colon cancer cells (13). Reduced mitochondrial O 2 consumption accounts for the degradation of HIF-1α in Nrf2-deficient cancer cells.…”

“…We next investigated the role of Nrf2 on reparative angiogenesis in the ischemic retina. Nrf2 has been found to play an important role in angiogenesis in other contexts, both with respect to vascular development (12) and pathologic setting, such as tumor angiogenesis (13,17). To determine the importance of Nrf2 in ischemic retina, we compared wild-type (Nrf2…”

“…Overexpression of p62/SQSM1 in KRas-induced tumor cells exhibits to increase pro-inflammatory responses through the Nrf2 and nuclear factor-kappa B (NF-κB) activation, thereby leading to tumor progression. These results indicate that p62 accumulation due to autophagy defect is strongly correlated with tumor development [17][18][19].…”

“…Thus, this transcription factor has emerged as a novel target for the prevention of colon cancer (Saw & Kong, 2011). However, stable RNAi-mediated knockdown of Nrf2 in human colon cancer cells suppressed tumor growth in mouse xenograft settings and colon tumor angiogenesis by inhibiting HypoxiaInduced Activation of HIF-1a (Kim et al, 2011). Thus, the role of Nfr2 in colon carcinogenesis still has to be explored.…”
