Novel Vaccination for Allergy through Gene Silencing of CD40 Using Small Interfering RNA

Abstract: Small interfering RNA (siRNA) is a potent means of inducing gene-specific silencing. Gene silencing strategies using siRNA have demonstrated therapeutic benefits in animal models of various diseases, and are currently in clinical trials. However, the utility of gene silencing as a treatment for allergic diseases has not yet been reported. In this study, we report a novel therapy for allergy through gene silencing of CD40, a critical costimulatory molecule and a key factor in allergic immune responses. Silencin… Show more

“…Mechanistically, calcitriol also has been shown through mediate effects through inhibition of NFkB (15). We have previously demonstrated that RNA interference may be evoked in DCs with short interfering RNA (siRNA) without induction of DC maturation or alteration of viability (16)(17)(18). These findings show that suppression of genes such as IL-12 p35, CD40, and RelB could endow DCs with the ability to inhibit immune responses both in vitro and in vivo (17,18).…”

These findings show that suppression of genes such as IL-12 p35, CD40, and RelB could endow DCs with the ability to inhibit immune responses both in vitro and in vivo (17,18). For example, administration of DCs pulsed with the model Ag OVA followed by subsequent silencing of IL-12 p35 led to a Th2 shift in Ag-specific recall response (16).…”

“…RNA interference is a posttranscriptional mechanism of gene silencing, in which 19-23 nucleotide double-stranded RNA duplexes promote specific endonucleolytic cleavage of mRNA targets called small interfering RNA (siRNA) as described in Raoul et al (2005) and Suzuki et al (2008). Gene silencing using siRNA is an extremely efficient, specific, long-lasting, and simple method for blocking the expression of target genes as described by Hill et al (2003).…”

“…The activation and differentiation of T cells play an important role in the pathogenesis of myocarditis. 1,40,41) First, we demonstrated that Th1-type cytokine production was markedly increased in the negative control group, and that this cytokine production was significantly suppressed in myocarditis rats after CD40 siRNA treatment. However, production of Th2-type cytokines was higher in the CD40 siRNA group compared with the negative control group.…”
