Attenuation of Experimental Autoimmune Myocarditis by si-RNA Mediated CD40 Silencing

Abstract: SummaryCD40 plays an important role in the pathogenesis of myocarditis, and inhibition of CD40 expression could be a promising treatment for myocarditis. In this study, we used an animal model, experimental autoimmune myocarditis (EAM), to investigate whether CD40 siRNA could be exploited for myocarditis therapy.Lewis rats were immunized with purified porcine cardiac myosin to induce EAM or were injected with phosphatebuffered saline (PBS) alone (PBS group), scrambled small interfering RNA (siRNA) (negative co… Show more

“…CD40 belongs to the TNF receptor superfamily and, as a costimulatory molecule, plays pivotal roles in the activation and differentiation of T cells . Besides, CD40 also plays a role in the development of myocarditis, rheumatoid arthritis, and autoimmune thyroiditis, and many autoimmune disorders . Previous research has established that suppression of CD40 expression could provide a good prospect for myocarditis treatment .…”

“…22 CD40, which has been found to be widely overexpressed in myocarditis, plays an important role in the pathogenesis of myocarditis and may become a potential target for the treatment of myocarditis. 23,24 Similarly, we discovered that CD40 was highly expressed in myocarditis patients, and silencing CD40 could inhibit the pro-inflammatory factors (TNF-α and IL-6) level and promoted the anti-inflammatory factor (IL-10) level. Importantly, CD40 was identified as a target of curcumin.…”
