Novel Targeted Anti-Tumor Nanoparticles Developed from Folic Acid-Modified 2-Deoxyglucose

Abstract: The glucose analog, 2-deoxyglucose (2-DG), specifically inhibits glycolysis of cancer cells and interferes with the growth of cancer cells. However, the excellent water solubility of 2-DG makes it difficult to be concentrated in tumor cells. In this study, a targeted nano-pharmacosome was developed with folic acid-modified 2-DG (FA-2-DG) by using amino ethanol as a cleavable linker. FA-2-DG was able to self-assemble, forming nano-particles with diameters of 10–30 nm. The biological effects were evaluated with … Show more

“…The clathrin/caveolin independent pathway also contributes to cell entry of metal oxide NPs. The, clathrin/caveolin independent pathway is important in the uptake of cellular fluids, hormones, and folic acid, and folic acid is of particular interest given its role in targeting nanomaterials to certain forms of solid tumors [ 112 , 113 ]. Macropinocytosis and micropinocytosis play minor roles in the cellular trafficking of metal oxides in most studies, and consequently they will not be considered further in this review.…”

“…By designing the ligands of the characteristic molecules of tumor cells on the surface of the nanoparticles, the specific binding between the nanoparticles and the tumor cells is promoted. In order to achieve targeted therapy of tumors, increase the concentration of drugs in the target area, thereby improving the utilization and efficacy of drugs and reducing adverse drug reactions [12,13]. Studies have shown that the PAH model pulmonary arteriole media smooth muscle cell surface glucose transporter-1 (GLUT-1) is highly expressed, aerobic glycolysis is enhanced, and 18 fluorodeoxyglucose (18F-FDG) uptake is increased [14].…”

“…The preparation of FA-2-DG has been reported previously [39]. The structure of FA-2-DG was confirmed with FTIR, nuclear magnetic resonance, and MS. All solvents and biochemical reagents were of analytical grade and were purchased from commercial sources (J&K Scientific, Beijing, China; Solarbio Science & Technology, Beijing, China).…”
