Aim: The objectives of this investigation were to develop gastroretentive floating bioadhesive drug delivery for simvastatin to increase gastric residence time and reduce the dose frequency. To explore Badams gum's sticking property in floating bioadhesive drug delivery of simvastatin. Materials and Methods: Floating bioadhesive tablet was formulated with Badam gum and hydroxypropyl methylcellulose (HPMC) K 15 M polymers, sodium bicarbonate, and citric acid as the gas generating agents to reduce floating lag time. Tablets were prepared by direct compression method. Optimization study was conducted using 3 2 factorial design. The concentration of polymers was considered as independent variables whereas swelling index, bioadhesive strength and % drug release at 10 h, of the tablets were utilized as dependent variables. Results and Discussions: Preformulation study suggests powders blends shows acceptable flow properties. Simvastatin floating-bioadhesive tablet found to be good without chipping, capping, and sticking. Comparing all the formulations, A5 optimized formulation exhibited 98.10 ± 2.10% of drug release in 12 h, floating lag time of 34 ± 3 s, with appropriate bioadhesive property, and total floating time of over 12 h. It was observed that increasing percentage of polymer in formulation the drug release decreased. Developed formulations were stable during stability studies. Conclusion: Based on these findings, it was concluded that Badam gum can be used as a bioadhesive as well as release retarding polymer for the floating bioadhesive dosage form of other drugs.

The aim of the present work was in vitro and in vivo evaluation of mucoadhesive tablets of rebapimide to prolong the gastric residence time after oral administration. The solubility of rebapimide was enhanced by kneading technique with that mixture formulations were prepared by using 3 3 full factorial designs to explore the effects of gum Kondagogu, gum Olibanum and Guar gum (as independent variables) on mucoadhesive strength, drug release and Ex vivo residence time (as dependent variables) was studied and published in the earlier research paper.In this investigation the formulated mucoadhesive tablets which was optimized through in vitro studies was selected and performed the in vivo studies on human volunteers. The drug-polymer interaction was also studied by conducting FTIR and DSC tests. The in vitro release kinetics studies reveal that all formulations fits well with zero order, followed by Korsmeyer-Peppas, Higuchi and the mechanism of drug release is erosion. After analysis of different evaluation parameters and drug release kinetics, formulation code F13 was selected as a promising formulation for delivery of rebapimide as a mucoadhesive gastroretentive tablet with best mucoadhesive strength and 99.34% drug release at 12 th hour. Radiological evidences suggest that, a formulated tablet was well adhered for >10 h in human stomach. The bioavailability studies of F13 containing rebapimide was carried out which exhibited increased pharmacokinetic parameters of Cmax (427.01±73), Tmax (4.00±1.23 h) and AUC 0-t (2242±18.24) as compared to marketed formulations which indicates improved bioavailability of formulations.

The aim of the present study was to develop and characterize a gastroretentive formulation for controlled drug release and to develop innovative gastro retentive formulation based on mucoadhesive patch systems using the solvent casting technique. Glipizide, an antidiabetic agent was used as model drug for formulating films. Mucoadhesive film was formulated using chitosan and HPMC K4M. PEG 400 was added as plasticizer in film preparation. 3 2 full factorial design was used to formulate the novel gastroretentive formulation. Amount of HPMC K4M(X1) and amount of chitosan (X2) was selected as independent variable while swelling index, folding endurance, mucoadhesion force and Q8 (% drug release after 8 h) was selected as dependent variables. The film with zigzag folding in the capsule was shown to unfold and swell under acidic conditions and provide controlled release of drug upto12 h in acidic medium. According to 3 2 full factorial design films, 9 batches were prepared and evaluated for surface pH, folding endurance, mucoadhesion force, drug content, in-vitro drug release etc. Surface pH of F1-F9 was in between 6.32 to 6.98. Thickness and % drug content for batch F1-F9 was found to be in between 0.236 mm to 0.289 mm and 95.42 % to 99.32 %, respectively. In-vitro drug release study of F1-F9 showed utmost 95 % drug release after 11 h. The results indicate that the dosage form is gastroretentive and can provide controlled release of drugs with narrow therapeutic window. Glipizide/ HPMCK4M / Chitosan (40:150:150) F8 was found to be optimized composition of mucoadhesive films that showed good swelling index, folding endurance, surface pH, mucoadhesion force, Q8 and % drug content.
