Novel PET Imaging of Atherosclerosis with 68Ga-Labeled NOTA-Neomannosylated Human Serum Albumin

Abstract: Activated macrophages take up 18 F-FDG via glucose transporters, so this compound is useful for atherosclerosis imaging by PET. However, 18 F-FDG application is limited for imaging of the heart and brain, in which glucose uptake is high, and in patients with aberrant glucose metabolism. The aims of this study were to confirm that mannosylated human serum albumin (MSA) specifically binds to the mannose receptor (MR) on macrophages and to test the feasibility of 68 Ga-labeled NOTA-MSA for PET imaging of atherosc… Show more

“…First, our study does not prove whether 68 Ga-NOTA-MSA is specific to imaging myocarditis or whether it targets various diseases associated with MR-positive macrophage infiltration. We think it would be the latter based on the findings that 68 Ga-NOTA-MSA can be used for tracking the sentinel lymph node 7,16 and also imaging atherosclerosis 40. It would be clinically interesting to expand our findings to other cardiovascular diseases as the importance of MR-positive macrophages is highlighted in various previous papers.…”

“…Several radioligands, including those targeting inducible nitric oxide synthase (21), folate receptor-b (22,23), CD206 (24,25), and LOX-1 (26)(27)(28), have been developed to visualize macrophages, and several of these have also been used to image acute inflammation. Though present ubiquitously, TSPO expression is elevated in immunologically activated leukocytes such as microglia and macrophages (29) and has been used as a molecular target for imaging of neuroinflammation (30)(31)(32)(33).…”

“…99m Tc-MSA, developed for SLN imaging 10 , was shown to bind to the mannose receptor on the macrophage surface 20 22 . Thus, 99m Tc-MSA may serve as an excellent agent for SLN imaging.…”
