Novel Macula-Like Virus Identified in Bombyx mori Cultured Cells

Abstract: We identified a novel, 6,513-bp-long RNA, termed Bombyx mori macula-like latent virus (BmMLV) RNA, which abundantly expressed in B. mori cultured BmN cells. BmMLV RNA potentially encodes two proteins, putative RNA replicase and coat protein, which share structural features and sequence similarities with those of a plant RNA virus, the genus Maculavirus. Northern blot analysis showed that two transcripts were expressed in BmN cells: a 6.5-kb-long RNA, which contains both putative RNA replicase and coat protein … Show more

“…mori larvae (F1 hybrid Kinshu × Showa) were reared as described previously (Katsuma et al, 2005). The Sf-9 and High Five cell lines were cultured at 27°C in TC-100 medium supplemented with 10% fetal bovine serum and Express Five medium (Invitrogen, Carlsbad, CA), respectively.…”

“…Two days after transfection, zeocin (final concentration, 500 mg/mL) was added to the medium. Two weeks after drug selection, the expression of FLAG-tagged Siwi and BmAgo3 was verified using the previously described Western blot analysis (Katsuma et al 2005) with anti-FLAG monoclonal (1:1000, Sigma), anti-Siwi (1:10,000), or antiBmAgo3 (1:5000) antibodies.…”

“…Infection typically causes a global shutoff of host protein synthesis and gene expression in insect cells beginning at around 12 to 18 h postinfection (p.i.) (hpi) (29,42,46). Conversely, particular host genes are induced or remain stably expressed until the late stage of baculovirus infection.…”
