Novel function of FAXDC2 in megakaryopoiesis

Abstract: FAXDC2 (fatty acid hydroxylase domain containing 2) is a member of the fatty acid hydroxylase superfamily. Given the important role of fatty acids in megakaryocytes, we have studied the role of this gene in the development of this lineage. Here we show that the expression of FAXDC2 is constantly elevated during megakaryocyte maturation. In contrast, FAXDC2 is significantly downregulated in acute myeloid leukemia and acute megakaryoblastic leukemia. Moreover, FAXDC2 overexpression promotes the differentiation o… Show more

“…However, we note that ATF4 may also be proviral (Caselli et al, 2012;Lee et al, 2018;Qian et al, 2012). Other low-TE genes include FAXDC2, STYX, SHOC2 which are involved in the Ras/Raf/Mitogen-activated protein kinase/ERK Kinase (MEK)/Extracellular-signal-Regulated Kinase (ERK) cascade (Jin et al, 2016;Reiterer et al, 2013;Rodriguez-Viciana et al, 2006). Importantly, it is commonly involved in viral infection, including that of coronaviruses (Kumar et al, 2018).…”

“…FAXDC2 belongs to the fatty acid hydroxylase superfamily that includes fatty acid and carotene hydroxylases and sterol desaturases, which are involved in cholesterol biosynthesis [27]. Specifically, fatty acid hydroxylase can alter the packing structures of membrane microdomains, and thus, regulates cellular function of membrane-associated proteins [28].…”

“…CMK cells treatment with Nocodazole (10 ng/ml), Methotrexate (2.5 nM), Etoposide (10 μM), and TPA (50 nM) was carried out as previously described [14,18,[33][34][35]. Human megakaryocyte culture experiments were performed by culturing CD34 + cells (purchased from Fred Hutchinson Cancer Research Center, USA) in StemSpam SFEM media (Stemcell Technologies, Vancouver, BC, Canada) supplemented with penicillin/streptomycin and lipids (40 mg/ ml) as well as megakaryocyte differentiation cytokine cocktail containing stem cell factor (100 ng/ml) and TPO (10 ng/ml) for 10 days [35,36]. Culturing mouse megakaryocytes from fetal liver cells was carried out as previously described [37].…”
