Novel fluoroimmunoassay for ovarian cancer biomarker CA-125

Abstract: Cancer antigen 125 (CA-125) is a glycoprotein biomarker that denotes the presence of ovarian and reproductive cancers in women, with serum concentrations of CA-125 greater than 35 U/ml considered indicative of potential malignancies. A fluorescent immunoassay recently developed in our laboratory employing the ALYGNSA antibody-orientation system has been used to measure CA-125 levels. This system displayed significantly increased sensitivity with a detection limit of 1.5 U/ml compared to that of a commercial CA… Show more

“…In this study, an assay was developed for the detection of the sECD-HER-2/neu protein utilizing the ALYGNSA system consisting of a protein biolinker (Protein G') and poly (methyl methacrylate) or PMMA, a thermoplastic polymer. The unique interaction of Protein G' with PMMA has been demonstrated to improve human IgG capture antibody alignment/orientation [12] and provide greater sensitivity in detection of cancer biomarkers CA-125 [13] and PSA [14].…”

“…This biomarker denotes the presence of ovarian and reproductive cancers in women, with serum concentrations of CA125 greater than 35 U/ml considered indicative of potential malignancies [3]. Interpretation of rising CA125 levels in diagnosed patients continues to be a topic of conversation among clinicians [4].…”

“…Considering the high importance and potential application in the early diagnosis of cancer, detection of tumor markers has received more and more attention [1923]. While some novel immunoassays have been developed [2433], a variety of techniques have been employed for the development of these assays. Table 2 summarizes some examples of immunoassays which have been developed for tumor markers analysis [3349].…”
