Novel discovery of Averrhoa bilimbi ethanolic leaf extract in the stimulation of brown fat differentiation program in combating diet-induced obesity

Abstract: Background Brown adipocytes are known to promote energy expenditure and limit weight gain to combat obesity. Averrhoa bilimbi , locally called belimbing buluh (DBB), is mainly used as an ethnomedicine in the treatment of metabolic disorders including diabetes mellitus, hypertension and obesity. The present study aims to investigate the browning activity on white adipocytes by A. bilimbi leaf extract and to evaluate the potential mechanisms. … Show more

“…Gene program commitments in myoblast-brown adipocyte transition were observed in bilimbi treated cells and the up-regulation of PGC-1α, UCP1 and PRDM16 proteins which influenced the cellular metabolism and mitochondrial function. 6 Our observations in this study showed that bilimbi n-Hex partition was able to induce a higher cellular response in signalling the co-development of brown adipocytes and myocytes than its crude extract. The induction of brown adipocyte differentiation indicates the presence of active phytocomponents that are responsible for the program.…”

“…[2][3][4][5] Its beneficial effects in brown adipogenesis activation, a phenomenon that combats obesity, has also been recently reported. 6 There are two main types of adipose tissue. White adipose tissue (WAT) is the primary site of fat storage and its amount increases in obesity.…”

“…Study by Fauziah et al (2018) showed that ethanol extract of A.bilimbi leaves a dose of 800 mg/kg BW can reduce total cholesterol and LDL levels in hypercholesterolemic mice. Meanwhile, in vitro research states that A.bilimbi leaves extract promoted adipogenesis in much the same way as a PPAR gamma agonist did (Lau et al, 2019). This study will continue the research of ethanol extract A.bilimbi leaves conducted by Fauziah et al, (2018), the ethyl acetate fraction of A.bilimbi leaves by Fauziah et al, (2020) and to determine the best dose of ethyl acetate fraction from A.bilimbi leaves, which have pharmacology activity on total cholesterol and triglycerides.…”

“…In addition, an HTS/HCS reader was also applied to the analysis of morphological changes in adipocytes without drug screening [41] . Lau et al used an HTS/HCS reader to analyze and quantify the expression of specific intracellular proteins in adipocytes [41] . It resolved the traditional dilemma of estimating the distribution and expression of intracellular proteins at the same time and may therefore indirectly replace immunoblotting.…”
