Novel biological function of sialic acid (N‐acetylneuraminic acid) as a hydrogen peroxide scavenger

Abstract: We have found that N-acetylneuraminic acid (NANA) consumes toxic hydrogen peroxide (H 2 O 2 ) under physiological conditions. Close investigation of this ¢nding revealed that NANA was oxidized by an equimolar amount of H 2 O 2 to provide its decarboxylated product, 4-(acetylamino)-2,4-dideoxy-D-glycero-D-galacto-octonic acid (ADOA). To date, there have been little data on this reaction, and its physiological signi¢cance has not been discussed. Examining the detoxi¢cation of H 2 O 2 in cultured cells with NANA,… Show more

“…Release of free monomeric sialic acid when it is cleaved from the sugar chain can detoxify H 2 O 2 (22). This reaction reduces the dangerous oxygen radicals, as H 2 O 2 and NANA are converted into H 2 O and nontoxic carboxylic acid (21,22).…”

“…Sialidase activity using the Amplex Red neuraminidase kit was tested in the total cell fraction (T), the extracellular fraction (Ex), and cell lysate (CL). * , P Ͻ 0.001. against oxidative damage (22). The relative significance of the P. gingivalis sialidase and O-sialoglycoprotease in oxidative stress resistance was evaluated by exposure of the isogenic defective mutants to hydrogen peroxide.…”

“…Presently, studies investigating the correlation between serum total sialic acid and the severity of atherosclerosis have presented conflicting results. It is noteworthy that recent studies have demonstrated that sialic acid could scavenge ROS 13, 14, 15 and may be applied as therapeutic option in some diseases 16, 17, 18.…”
